Human Factor XIII is a tetramer composed of two pairs of chains held together by noncovalent bonds. After activation of the zymogen via Thrombin to its active enzyme form, Factor XIIIa is responsible for catalyzing the formation of covalent bridges between fibrin units to increase the elasticity of the clot network. The resulting cross-linked fibrin is very insoluble and resistant to lysis.

The human factor XIII is ≥94% as judged by SDS-PAGE. One plasma equivalent unit (PEU) is equal to ~108 Loewy units. The above protein was purified from Human plasma units that test negative for antibodies to HBc, HCV, HIV 1/2, HTLV-I/II, and non reactive for HBsAg, HIV-1 RNA, HCV RNA, HBV DNA and Syphilis by FDA approved tests. Donors are screened for CJD.

生化机理

Factor XIII is activated by thrombin and calcium ion to a transglutaminase that catalyzes the formation of gamma-glutamyl-epsilon-lysine cross-links between fibrin chains, thus stabilizing the fibrin clot. Also cross-link alpha-2-plasmin inhibitor, or fibronectin, to the alpha chains of fibrin. The B chain of factor XIII is not catalytically active, but is thought to stabilize the A subunits and regulate the rate of transglutaminase formation by thrombin.
