Recombinant Rat IL-12 Protein (rp167051) - Protein Bioactivity
Measured in a cell proliferation assay using PHA-activated mouse splenocytes. The ED50 for this effect is 0.025-0.25 ng/mL.
Recombinant Rat IL-12 Protein (rp167051) - SDS-PAGE
2 μg/lane of Recombinant Rat IL-12 Protein was resolved with SDS-PAGE under reducing (R) and non-reducing (N) conditions and visualized by Coomassie® Blue staining, showing the bands at 26 & 46-52 kDa under reducing condition and 40-48 & 65-72 kDa under non-reducing condition.
The core structural feature of IL‑12 lies in its heterodimeric form, composed of the p35 and p40 subunits covalently linked by disulfide bonds. The p35 subunit adopts an α‑helical structure and is unique to IL‑12, whereas the p40 subunit contains fibronectin type III and Ig C2‑like domains, showing high topological homology to type I cytokine receptors. In addition, the p40 subunit exhibits remarkable structural diversity and sharing capacity: it can not only pair with p35 to form IL‑12, but also exist as a monomer, homodimer, or heterodimer with p19 to form IL‑23.
Meanwhile, the potential N‑glycosylation sites on the p35 subunit provide a structural basis for its post‑translational modifications.
