生化机理
Human Complement 5a (C5a) is an enzymatically generated glycoprotein that belongs to a family of structurally and functionally related proteins known as anaphylatoxins. C5a is a 74 amino acid (aa) peptide that is created by the activity of C5a convertase on the C5 alpha -chain. Human C5a has four alpha -helices plus three intrachain disulfide bonds that create a triple loop structure. In serum, proteolytic processing removes the C‑terminal arginine, creating a low activity C5adesArg74 molecule. Human C5a is 60% and 54% aa identical to mouse and rat C5a, respectively. C5a binds to a signaling G-protein coupled receptor (C5aR/CD88) and a nonsignaling GPCR termed C5L2. Activation of Cd88 results in neutrophil chemotaxis and endothelial cell activation. It also triggers an oxidative burst in macrophages and neutrophils, and induces release of histamine in basophils and mast cells.
Post-translational: C5 precursor is first processed by the removal of 4 basic residues, forming two chains, beta and alpha, linked by a disulfide bond (PubMed:106884). During activation of the complement systems, the alpha chain is cleaved into C5a and C5b by the C5 convertase: C5b stays linked to the beta chain, while C5a is released in the plasma (PubMed:12878586, PubMed:18204047, PubMed:30643019, PubMed:624565). The alpha chain is cleaved by the serine protease complement C2b component of the C5 convertase to generate C5a and C5b following activation by the classical, lectin and GZMK complement systems (PubMed:12878586, PubMed:18204047).The alpha chain is cleaved by CFB component of the C5 convertase to generate C5a and C5b following activation by the alternative complement system (PubMed:30643019, PubMed:624565, PubMed:6554279).
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Reconstitute at 0.5 mg/mL in sterile distilled water. Stock solutions should be apportioned into working aliquots and stored at ≤ -20 °C. Further dilutions should be made in appropriate buffered solutions.
