生化机理
Centromere Protein U, also known as CENPU is a specialized chromatin domain, present throughout the cell cycle. CENPU performs as a platform on which the transient assembly of the kinetochore takes place during mitosis. CENPU plays a major role in the correct PLK1 localization to the mitotic kinetochores. A scaffold protein is responsible for the first recruitment and maintenance of the kinetochore PLK1 population until its degradation. CENPU is also implicated in transcriptional repression.
Post-translational: Phosphorylated by PLK1 at Thr-78, creating a self-tethering site that specifically interacts with the polo-box domain of PLK1.

3 μg/lane of Recombinant Human CENPU Protein was resolved with SDS-PAGE under reducing (R) and non-reducing (N) conditions and visualized by Coomassie® Blue staining, showing the bands at 32.7 kDa under reducing conditions and 32.8 & 69.5 kDa under non-reducing conditions.
CENPU plays a central role in kinetochore assembly, mitotic progression, and chromosome segregation. Its amino acid region 147-418 is likely to contain coiled-coil domains or other protein-protein interaction interfaces. Such domains tend to form dimers or multimers when expressed in prokaryotic systems (Uniprot ID: Q71F23).
