Pdx1 C terminus-interacting factor-1 (Pcif1, also known as SPOP) is a nuclear protein that inhibits Pdx1 transactivation. PCIF1 is a TRAF and POZ domain-containing nuclear factor that interacts with and inhibits transactivation of pancreatic homeodomain transcription factor PDX-1. PCIF1 is composed of 704 amino acids. The WW domain of PCIF1 can directly and preferentially bind to the phosphorylated carboxy-terminal domain (CTD) compared to the unphosphorylated CTD. PCIF1 binds to the hyperphosphorylated RNAP II (RNAP IIO) in vitro and in vivo. Double immunofluorescence labeling in HeLa cells demonstrated that PCIF1 and endogenous RNAP IIO are co-localized in the cell nucleus. Thus, PCIF1 may play a role in mRNA synthesis by modulating RNAP IIO activity.
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Reconstitute at 1.0 mg/mL in sterile distilled water. Stock solutions should be apportioned into working aliquots and stored at ≤ -20 °C. Further dilutions should be made in appropriate buffered solutions.

Recombinant Human PCIF1 Protein (rp217615) - SDS-PAGE 3 μg/lane of Recombinant Human SOD1/Superoxide Dismutase 1 Protein was resolved with SDS-PAGE under reducing (R) and non-reducing (N) conditions and visualized by Coomassie® Blue staining, showing a band at 26.8 kDa under reducing conditions and 27.0 & 55.0 kDa under non-reducing conditions. The WW domain at PCIF1’s N-terminus mediates its dimerization through self-association, confirming the WW domain’s role in PCIF1 oligomerization (PMID: 18562638).
