胰蛋白酶选择性水解变性蛋白质中由赖氨酸或精氨酸的羧基所构成的肽链，是一种重要的消化酶。此外，胰蛋白酶还广泛应用于脓胸、血胸、外科炎症、溃疡、创伤性损伤等所产生的局部水肿、血肿及脓肿等的辅助治疗。在该试剂盒中，胰蛋白酶催化水解 TAME 的酯键，释放出的游离羧基与反应体系中的氢氧化钠发生中和反应，导致溶液的 pH 值降低，以苯酚红为指示剂，测定溶液在 555 nm 处吸收值的变化，可以快速测得胰蛋白酶活性数据。胰蛋白酶在一定范围内与 555 nm 处吸收值的降低呈良好的线性关系。
Trypsin selectively hydrolyzes peptide bonds formed by the carboxyl groups of lysine or arginine in denatured proteins, making it an important digestive enzyme. Additionally, trypsin is widely used as an adjunct therapy for conditions such as local edema, hematomas, and abscesses resulting from empyema, hemothorax, surgical inflammation, ulcers, traumatic injuries, and similar cases. In this assay kit, trypsin catalyzes the hydrolysis of the ester bond in TAME, releasing free carboxyl groups. These groups undergo a neutralization reaction with sodium hydroxide present in the reaction system, leading to a decrease in the pH of the solution. Using phenol red as an indicator, the change in absorbance at 555 nm is measured, enabling rapid determination of trypsin activity. Within a certain range, trypsin activity exhibits a well-defined linear relationship with the decrease in absorbance at 555 nm.
