Thiol groups in living organisms primarily consist of glutathione thiols and protein thiols. The former not only helps repair oxidatively damaged proteins but is also involved in scavenging reactive oxygen species, while the latter plays a crucial role in maintaining protein conformation. By measuring the total thiol content and glutathione content, the protein thiol content can be determined indirectly. Thiol groups react with 5,5’-dithio-bis-nitrobenzoic acid (DTNB), generating a yellow-colored compound which exhibits a maximum absorption peak at 412 nm.
