Vero Serum-Free Medium is a serum‑free basal medium that contains no proteins or any animal‑derived components. It is specifically developed for the Vero cell line and can be used for adherent static culture as well as microcarrier‑based suspension large‑scale culture of Vero cells. It supports high‑density cell growth and maintains high cell viability, and is suitable for the research, development and production of influenza vaccines and related products.
Applications
Vero Serum-Free Medium can be used for the recovery, subculture, adherent static culture, and microcarrier‑based suspension large‑scale culture of Vero cells. This basal medium is intended for research use and for cell‑culture‑based large‑scale production of biological products, but it is not for direct use in humans or as a drug.
Cell Recovery
1.Pre‑warm Vero Serum-Free Medium in a 37 °C water bath.
2.Spray the medium bottle with 75% alcohol and place it in a biological safety cabinet.
3.Thaw one vial of cells by gently shaking it in a 37 °C water bath for 1 minute.
4.Gently transfer the cells from the cryovial into a centrifuge tube containing 10 mL of pre‑warmed Vero Serum-Free Medium.
5.Centrifuge at 150–300 g (approximately 800–1000 rpm) for 5 minutes. Discard the supernatant and resuspend the cell pellet in 10 mL of fresh pre‑warmed medium. Adjust the viable cell density to 2–4 × 10⁴ cells/mL.
6.Take 0.5 mL of the cell suspension and measure the viable cell density (×10⁶ cells/mL) and cell viability (%) using a cell counter.
7.If cell viability is > 90%, place the cells under the specified culture conditions for further culture.
Cell Subculture
(Using a T75 flask as an example)
1.Pre‑warm Vero Serum-Free Medium at 37 °C for 20–30 minutes.
2.Use cells in the mid‑logarithmic growth phase with a viability ≥ 90% for subculture.
3.Remove the medium from the flask to be subcultured, rinse the cell layer once with 7–10 mL of DPBS, discard the wash, and repeat once more.
4.Add 1 mL of recombinant trypsin digestion solution to cover the entire growth surface and incubate at room temperature for 3–5 minutes.
5.When the cells detach from the flask wall, tap the flask gently, then add the pre‑warmed mixture (1 mL of 0.5% trypsin inhibitor + 9 mL of Vero Serum-Free Medium, total 10 mL) to pipette and disperse the cells. Collect the cell suspension and centrifuge at 300 g for 5–10 minutes at room temperature.
6.Discard the supernatant and resuspend the cell pellet in Vero Serum-Free Medium.
7.Calculate the volume of seed cell suspension needed based on a seeding density of 2–4 × 10⁴ cells/cm² and the subculture volume.
8.Aseptically transfer the required volume of seed suspension into a T75 flask containing the required volume of pre‑warmed Vero Serum-Free Medium.
9.Subculture every 3–5 days.
Cell Adaptation
1.For cells that can be directly adapted, they may be inoculated directly from serum‑containing medium into Vero Serum-Free Medium. The seeding density should follow the subculture procedure, but may need to be adjusted according to the specific situation.
2.Continue subculturing the cells until they grow stably.
3.A recommended seeding density for subculture is 2–4 × 10⁴ cells/cm², with subculture every 4–6 days. Typically, cells become fully adapted after 3 passages.
Cell Cryopreservation
1.Prepare cells in the mid‑logarithmic growth phase with viability > 90% and in good condition.
2.Determine the viable cell density and calculate the required volume of cryopreservation medium so that the final viable cell density is > 1 × 10⁷ cells/mL.
3.Prepare the required cryopreservation medium: 90% Vero Serum-Free Medium + 10% DMSO, and store at 4 °C.
4.Prepare a single‑cell suspension following the cell digestion procedure.
5.Centrifuge at 300 g for 5 minutes, discard the supernatant, and resuspend the cells in the cryopreservation medium.
6.Aliquot the suspension into suitable cryovials according to project requirements.
7.Cool the cryovials using a standard controlled‑rate freezing protocol (automatic or manual, at approximately 1 °C per minute).
8.Transfer the cryovials to liquid nitrogen for long‑term storage.
