Protective Effects of Four Structurally Distinct Sanshools Ameliorate Dextran Sodium Sulfate-Induced Ulcerative Colitis by Restoring Intestinal Barrier Function and Modulating the Gut Microbiota

Zhaojun Chen,Hui Wang,Lulin Tan,Xiong Liu

Hydroxy-α-sanshool (HAS), hydroxy-β-sanshool (HBS), hydroxy-γ-sanshool (HRS), and γ-sanshool (RS) are the key components from theZanthoxylumgenus, processing a range of pharmacological activities. The present study investigated the protective capacities of four sanshools on a dextran sulfate sodium (DSS)-induced model of ulcerative colitis (UC). The results showed that sanshool administration alleviated the colitis symptoms by reducing body weight loss and disease activity index (DAI) score, increasing the colon length, and improving colonic injury and the change in immune organ weight. Furthermore, sanshools enhanced the antioxidant enzyme activities, and RS exhibited the lowest effect on the improvement in total antioxidative capacity (T-AOC) and antioxidant abilities compared to the other three sanshools. The p65 nuclear factor κB (p65 NFκB) signaling pathway was inhibited to prevent hyperactivation and decreased the production of inflammatory factors. The gut barrier function in DSS-induced mice was restored by increasing goblet cell number and levels of tight junction proteins (zonula occludens-1, occludin, and claudin-1), and the levels of protein in HAS and HRS groups were higher than that in the HBS group, significantly. The analysis of gut microbiota suggested that sanshool administration significantly boosted the abundance ofLachnospiraceae,Muribaculaceae,Oscillospiraceae, andAlistipesand reduced the level ofBuchnerain colitis mice. Collectively, the sanshool treatment could ameliorate colitis by resisting colon injury and regulating intestinal barrier dysfunction and gut microbiota dysbiosis; meanwhile, HRS and HAS have better improvement effects. Keywords:sanshool;colitis;intestinal barrier;p65;intestinal flora

Intestinal absorption and distribution of naringin, hesperidin, and their metabolites in mice

Xiao Guo,Kaikai Li,Ailing Guo,Erhu Li

In this study, an in vivo animal experiment was performed, to investigate the intestinal absorption and distribution of naringin, hesperidin, and their metabolites by using UPLC-ESI-MS/MS and HPLC-PDA analysis. The bioavailability of naringin or hesperidin in duodenum, jejunum, ileum, cecum, and colon was calculated. A total of 7 and 13 metabolites were found and tentatively identified in intestinal tissue   after orally administered naringin and hesperidin in mice, respectively. Sulfates of naringin could be absorbed by ileum and cecum, de-glycosylation metabolites could be absorbed by duodenum, and phenolic acids could be absorbed by the cecum and colon. Sulfates of hesperidin could be absorbed by duodenum and ileum, de-glycosylation metabolites could be absorbed by cecum, and phenolic acids could be absorbed by ileum, cecum and colon. Jejunum did not absorb metabolites of naringin or hesperidin. Results could be beneficial for understanding the intestinal bioactivities of naringin and hesperidin.

Bletilla striata Oligosaccharides Improve Ulcerative Colitis by Regulating Gut Microbiota and Intestinal Metabolites in Dextran Sulfate Sodium-Induced Mice.

Tianxiang Zhu,Baifei Hu,Cheng Ye,Haiming Hu,Mingzhu Yin,Zhigang Zhang,Shuiqing Li,Yanju Liu,Hongtao Liu

This study aimed to elucidate the mechanism of Bletilla striata oligosaccharides (BO) in the treatment of ulcerative colitis (UC). A UC mouse model was induced by 3% Dextran sodium sulfate (DSS), and BO (200 mg/kg/d) were administered for intervention. The results show that BO effectively inhibited the release of intestinal inflammatory cytokines such as IL-6, TNF-α, and IL-1β. Also, BO profoundly elevated the secretion of mucins and the expression of tight junction (TJ) proteins to attenuate dysfunction of the intestinal barrier. The 16S rDNA sequencing and liquid chromatography/gas chromatography-mass spectrometer (LC/GC-MS) analysis of mouse feces revealed that BO regulated the disturbance of gut microbiota and intestinal metabolites. By using the in vitro fermentation broth of BO and gut microbiota-depleted mice treated with antibiotics, we confirmed the protection of BO against UC. In conclusion, BO played a role in improving UC by modulating gut microbial composition and intestinal metabolites, which provided new therapeutic strategies for UC treatment. Keywords: Bletilla striata oligosaccharides, ulcerative colitis, gut microbiota, intestinal metabolites, intestinal barrier

Effect of Piper nigrum essential oil in dextran sulfate sodium (DSS)-induced colitis and its potential mechanisms

Yeye Hu,Ziliang He,Ji Zhang,Chaohua Zhang,Yanting Wang,Wei Zhang,Fenglun Zhang,Weiming Zhang,Fenglin Gu,Weicheng Hu

Background Piper nigrum essential oil (PnEO) possesses pleasant aroma, unique flavor, and various bioactivities; however, its role against colitis remains unclear. Purpose In this study, we investigated the role of PnEO in relieving colitis and explored its potential mechanisms in a mouse model of dextran sulfate sodium (DSS)–induced colitis. Methods Initially, we identified and quantified the components of PnEO by gas chromatography-mass spectrometry (GC–MS). Subsequently, we investigated the protective role of PnEO (50 and 200 mg/kg) in DSS-induced colitis in mice by evaluating disease activity index (DAI) scores and colon length, and performing histological analyses. Eyeball blood was collected and cytokines were determined using ELISA kits. The anti-inflammatory mechanisms of PnEO were analyzed by western blot (WB) and immunohistochemistry (IHC). The intestinal barrier function was evaluated according to tight junction (TJ) protein mRNA levels. We used 16S rRNA gene sequencing to analyze the intestinal microflora of mouse cecal contents. Results Supplementation with PnEO (50 and 200 mg/kg) increased colon length and improved colon histopathology . PnEO regulated inflammatory responses by downregulating TLR4/MAPKs activation, thereby reducing the release of cytokines and mediators. Moreover, it also protected the intestinal barrier through enhancing the expression of claudin-1, claudin-3, occludin , ZO-1, and mucin 2 . 16S rRNA gene sequencing revealed that PnEO (200 mg/kg) decreased the abundance of Akkermansia in the gut microbiome . Conclusion PnEO treatment (50 and 200 mg/kg) relieved DSS-induced colitis by inhibiting TLR4/MAPK pathway and protecting intestinal barrier, and high-dose PnEO exhibited better effects. Moreover, PnEO (200 mg/kg) regulated key compositions of the gut microbiome, which indicated that it had therapeutic potential for sustaining gut health to lower the risk of colitis.

Structural Characterization of Morchella esculenta Polysaccharides and Its Ability to Modulate Intestinal Barrier and Intestinal Microbiota in Dextran Sulfate Sodium-Induced Colitis Mice

Shutong Chen,Bo Teng,Wancong Zhang,Jude Juventus Aweya,Kit-Leong Cheong

Morchella esculenta polysaccharides (MEPs) are known to have multiple bioactive properties, including immunomodulatory, anticancer, antioxidant, anti-inflammatory, etc. In this study, we assessed the impact of MEPs on dextran sulfate sodium (DSS)-induced colitis in mice, focusing on intestinal barrier and microbiota modulation. Using NMR analysis, MEPs were found to predominantly consist of 1,4,6- α - d -Glc p and 1,4- α - d -Glc p . MEPs were able to significantly alleviate weight loss, reduce colon length shortening, and mitigate colon pathology in mice. Notably, MEPs suppressed elevated pro-inflammatory cytokines levels but boosted anti-inflammatory cytokine levels. Besides, MEPs modulated gut microbiota, enhancing microbial diversity and promoting homeostasis compared to untreated. Furthermore, there was a higher relative abundance of beneficial bacteria, indicating that MEPs could enhance gut microbiota composition. Collectively, MEPs have promising therapeutic potential in preventing colitis, and therefore could be developed as a novel nutraceutical strategy.

NVP-AUY922 relieves radiation-induced intestinal injury via regulating EPHX1

Wenzhe Kong,Xudong Ding,Zhaoyu Wang,Lu Lu,Saijun Fan

As a common side effect of radiotherapy, radiation-induced intestinal injury (RIII) greatly affects the prognosis of patients and the efficacy of radiotherapy. Current therapeutic strategies for RIII are still very limited. Thus, the identification of effective radioprotective agents is of great importance. NVP-AUY922 is an HSP90 inhibitor with favorable anti-inflammatory and antioxidant activities. It has been proven to mitigate radiation-induced lung injury. However, its effects on the alleviation of RIII remain unclear. In this study, our data indicated that NVP-AUY922 remarkably increased the survival rate after radiation exposure. NVP-AUY922 treatment could enhance the viability of intestinal stem cells (ISCs) and promote the recovery of the small intestine. In addition, it also inhibited intestinal inflammation and reshaped the gut microbiota structure. We found that the radioprotective effect of NVP-AUY922 is partially dependent on EPHX1. In addition, NVP-AUY922 could attenuate dextran sulfate sodium (DSS)-induced colitis and promote intestinal barrier recovery. Thus, our results suggest that NVP-AUY922 contributes to the amelioration of intestinal injury after radiation exposure, which offers a new approach for the prevention of RIII.

Piperine Improves DSS-Induced Colitis in Mice via Inhibition of Inflammation and Modulation of Gut Microbiota

Yeye Hu,Yanting Wang,Haoyang Gao,Guigui Yang,Jing Xie,Ziliang He,Shenghui Lv,Fenglin Gu,Chaoqing Huang,Weicheng Hu

Inflammatory bowel disease (IBD) is a global health concern with limited therapeutic options. Previous studies have demonstrated that piperine exhibited anti-inflammatory effects both in vitro and in vivo. However, its potential to ameliorate colitis in mice through modulation of gut microbiota has not been explored. This study aimed to investigate the role of gut microbiota in the protective effects of piperine against colitis using a dextran sulfate sodium (DSS)-induced mouse model. Mice were administered piperine (12.5 and 25 mg/kg) prior to DSS exposure. Fecal microbiota transplantation (FMT) was then performed, after which we evaluated colitis symptoms, inflammation levels, and intestinal barrier function. Subsequently, 16S rDNA-based high-throughput sequencing was employed to analyze the microbial composition of the mouse cecal contents. Piperine administration increased the colon length, decreased the spleen index, and improved colon histopathology. Furthermore, piperine modulated inflammatory responses by inhibiting NF-κB signaling, thereby reducing the release of pro-inflammatory cytokines and mediators. It also enhanced intestinal barrier integrity by increasing the expression of claudin-1, claudin-3, ZO-1, occludin, and mucin 2. Notably, the 16S rDNA sequencing results revealed that piperine increased the abundance of Dubosiella in the gut. Piperine effectively protected mice from DSS-induced colitis, suppressed inflammation, and improved poor intestinal barrier function. It reshaped the intestinal microbiota, ultimately alleviating DSS-induced colitis in mice. Our research highlighted the significant role of gut microbiota in the piperine-mediated alleviation of intestinal damage and suggested its therapeutic potential for promoting gut health and reducing the risk of colitis.

Flammulina velutipes polysaccharide counteracts cadmium-induced gut injury in mice via modulating gut inflammation, gut microbiota and intestinal barrier

Rili Hao,Xing Zhou,Xinyue Zhao,Xiaqing Lv,Xiangyang Zhu,NaNa Gao,Yang Jiang,Maoyu Wu,Dongxiao Sun-Waterhouse,Dapeng Li

Cadmium (Cd), as Group I carcinogen, can induce damage to various organs including the gut. It is of great importance to meet the rising demand for effective therapies against Cd-induced damage and investigate the mechanism. Flammulina velutipes is a popular edible mushroom, despite the well-known health benefits of Flammulina velutipes , little is known about the effect of its polysaccharide (FVP) against CdCl 2 -intestinal injury. In this study, a FVP (uronic acid, 5.10 %; degree of methylation, 41.24 %) was produced via hot water extraction (85 °C) and ethanol precipitation. The FVP contained eight major monosaccharides and exhibited good thermal stability at temperatures lower than 139.73 °C. FVP (100 mg/kg b. w., gavage for 4 weeks) alleviated CdCl 2 (1.5 mg/kg b. w., gavage for 4 weeks)-induced intestinal inflammation and apoptosis, intestinal permeability alteration and intestinal barrier disruption. FVP increased the abundance of Bacteroides , whilst decreasing the abundance of Desulfovibrionales and Clostridium . FVP also restored the levels of short-chain fatty acids (SCFAs), including acetic, propionic, isobutyric, butyric, isovaleric and valeric acids. Correlation analysis indicated the interplays among the FVP, gut microbes, SCFAs, intestinal barrier/cells and gut inflammation. FVP enhances the metabolic functions of gut microbiota via functional pathways analyzed by KEGG database. Furthermore, gut microbial transplantation of FVP + CdCl 2 group mice partially alleviated CdCl 2 caused-gut damage. Thus, FVP may be an effective therapeutic agent against CdCl 2 -induced gut damage via SCFA-mediated regulation of intestinal inflammation and gut microbiota-related energy metabolism. This study may open a new avenue for developing alternative strategies to prevent CdCl 2 -caused injury.

Polystyrene microplastics exacerbate experimental colitis in mice tightly associated with the occurrence of hepatic inflammation

Ting Luo,Dou Wang,Yao Zhao,Xinfang Li,Guiling Yang,Yuanxiang Jin

The potential health effects of microplastics (MPs) have become a public concern due to their ubiquitousness in the environment and life. Numerous studies have demonstrated that a high dose of MPs can adversely affect gastrointestinal health. However, few studies have focused on the impact of microplastics on patients' health with respect to gastrointestinal diseases. Inflammatory bowel disease (IBD) has emerged as a global disease with a rapidly increasing incidence. IBD, a specific gastrointestinal illness characterized by acute, chronic inflammation and intestinal barrier dysfunction, might increase sensitivity to MPs exposure. Herein, we investigated the impact and mechanism of PS-MPs on dextran sodium sulfate (DSS)-induced colitis. The results demonstrated that gavage with PS-MPs alone caused minimal effects on the intestinal barrier and liver status of mice. For mice with colitis, additional PS-MPs exposure caused a shorter colon length, aggravated histopathological damage and inflammation, reduced mucus secretion, and increased the colon permeability. Furthermore, PS-MPs exposure also increased the risk of secondary liver injury associated with inflammatory cell infiltration. These findings provide more histopathological evidence and suggest a need for more research on the health risk of MPs for sensitive individuals.

New pathway ameliorating ulcerative colitis: focus on Roseburia intestinalis and the gut-brain axis

Fenghua Xu,Yi Cheng,Guangcong Ruan,Liqin Fan,Yuting Tian,Zhifeng Xiao,Dongfeng Chen,Yanling Wei

Background: The community of gut microbes is a key factor controlling the intestinal barrier that communicates with the nervous system through the gut-brain axis. Based on our clinical data showing that populations of Roseburia intestinalis are dramatically decreased in the gut of patients with ulcerative colitis, we studied the efficacy of a strain belonging to this species in the context of colitis and stress using animal models. Methods: Dextran sulfate sodium was used to induce colitis in rats, which then underwent an enema with R. intestinalis as a treatment. The disease activity index, fecal changes and body weight of rats were recorded to evaluate colitis, while histological and immunohistochemical analyses were carried out to examine colon function, and 16S rRNA sequencing was performed to evaluate the gut microbiota change. Behavioral assays and immunohistochemical staining of brain were performed to assess the effect of R. intestinalis on the gut-brain axis. Results: Colitis-related symptoms in rats were significantly relieved after R. intestinalis enema, and the stool traits and colon length of rats were significantly recovered after treatment. The gut epithelial integrity and intestinal barrier were restored in treated rats, as evidenced by the higher expression of Zo-1 in colon tissues, accompanied by the restoration of gut microbiota. Meanwhile, depressive-like behaviors of rats were reduced after treatment, and laboratory experiments on neuronal cells also showed that IL-6, IL-7 and 5-HT were downregulated by R. intestinalis treatment in both serum and brain tissue, while Iba-1 expression was reduced in treated rats. Conclusions: The administration of R. intestinalis contributes to restoration of the gut microbiota, promoting colon repair and the recovery of gastrointestinal function. These alterations are accompanied by the relief of depressive-like behaviors through a process modulated by the neuronal network and the regulation of inflammation by the gut-brain axis.

Canthaxanthin improves dextran sulfate sodium-induced chronic colitis by modulating intestinal barrier and gut microbiota

Xiaoxiang Chen,Yan Chen,Zhichao Chen,Jiayao He,Xiahao Sun,Zuisu Yang

This study explored the beneficial effects of canthaxanthin (CX) on dextran sulfate sodium (DSS)-induced chronic colitis in mice and its underlying mechanisms. Oral administration of CX alleviated colitis severity, as demonstrated by reduced weight loss, diarrhea, hematochezia, colon shortening, and pathological colon injury. Improvements in intestinal permeability and inflammation were indicated by significantly decreased levels of permeability markers and pro-inflammatory cytokines. Moreover, CX mitigated mucus barrier dysfunction by enhancing goblet cell proliferation and mucin-2 secretion. CX reinforced the epithelial barrier by preserving tight junction function and preventing excessive apoptosis of colonic epithelial cells. Furthermore, CX modulated DSS-induced gut microbiota dysbiosis by reshaping the diversity and structure of both bacterial and fungal communities, increasing the abundance of beneficial microbes while reducing the prevalence of harmful microbes. To conclude, CX can effectively alleviate chronic colitis via modulating intestinal barrier function and maintaining microbiome homeostasis, providing a potential dietary therapy for UC.

Chemical Composition of Cynanchum auriculatum Royle Ex Wight and Its Potential Role in Ameliorating Colitis

Sichen Li,Yuning Sun,Huihui Peng,Ruiqiang You,Fuqing Bai,Dan Chen,Mohamed Abdin,Chuanyi Peng,Xiang Li,Huimei Cai,Guijie Chen

Cynanchum auriculatum Royle ex Wight, commonly known as “Baishouwu,” has been traditionally used in China for its medicinal and dietary benefits. Despite its long history of use, the potential therapeutic effects of C. auriculatum in the treatment of colitis have not been fully investigated. This study aims to evaluate the effects of the water extract of C. auriculatum root on colitis and elucidate its potential mechanisms of action. The water extract of C. auriculatum root (CW) was prepared and characterized using UPLC-Q-TOF-MS, identifying thirty-two distinct compounds, including saponins, organic acids, fatty acid derivatives, and alkaloids. The therapeutic efficacy of CW was assessed in a colitis mouse model. CW significantly alleviated colitis symptoms, evidenced by increased colon length, reduced disease activity indices, and decreased colon tissue damage. CW reduced colonic inflammatory cytokine production and enhanced the expression of tight junction proteins, including claudin-1, occludin, and ZO-1, thereby strengthening intestinal barrier integrity. Additionally, CW modulated the gut microbiota by increasing microbial diversity, promoting beneficial Lactobacillus growth, reducing pathogenic Pseudomonas levels, and enhancing short-chain fatty acid production. The results suggest that CW exhibits significant therapeutic potential in the management of colitis by attenuating inflammation, restoring gut barrier function, and modulating the gut microbiota. These findings provide a basis for further exploration of C. auriculatum as a functional food for prevention and treatment of colitis.
