Biochemical responses of freshwater microalgae Chlorella sorokiniana to combined exposure of Zn(Ⅱ) and estrone with simultaneous pollutants removal

Dongyang Liu,Hanzhi Wang,Yue Teng,Qirui Wu,Chunming Tang,Xinxin Gao,Chaoqi Chen,Liandong Zhu

With the development of livestock industry , contaminants such as divalent zinc ions (Zn (Ⅱ)) and estrone are often simultaneously detected in livestock wastewater. Nevertheless, the combined toxicity of these two pollutants on microalgae is still unclear. Moreover, microalgae have the potential for biosorption and bioaccumulation of heavy metals and organic compounds. Thus, this study investigated the joint effects of Zn (Ⅱ) and estrone on microalgae Chlorella sorokiniana , in terms of growth, photosynthetic activity and biomolecules , as well as pollutants removal by algae. Interestingly, a low Zn (Ⅱ) concentration promoted C . sorokiniana growth and photosynthetic activity, while the high concentration experienced inhibition. As the increase of estrone concentration, chlorophyll a content increased continuously to resist the environmental stress. Concurrently, the secretion of extracellular polysaccharides and proteins by algae increased with exposure to Zn (Ⅱ) and estrone, reducing toxicity of pollutants to microalgae. Reactive oxygen species and superoxide dismutase activity increased as the increase of pollutant concentration after 96 h cultivation, but high pollutant concentrations resulted in damage of cells, as proved by increased MDA content. Additionally, C. sorokiniana displayed remarkable removal efficiency for Zn (Ⅱ) and estrone, reaching up to 86.14% and 84.96% respectively. The study provides insights into the biochemical responses of microalgae to pollutants and highlights the potential of microalgae in pollutants removal.

Adsorption of estrone with few-layered boron nitride nanosheets: Kinetics, thermodynamics and mechanism

Guohong Liu,Zhaohan Zhang,Chen Yan,Yang Wang,Xuerui Ma,Peng Gao,Yujie Feng

Boron nitride Nanosheets (BNNSs) was fabricated with a method of heating the mixture of boric acid and urea in N 2 atmosphere and used to remove estrone (E 1 ) from water. The obtained BNNSs exhibited a higher surface area of 896 m 2 /g, a large pore volume of 0.76 cm 3 /g, and only few layers (0.398 nm) with the boric acid and urea ratio of 1:80. The layer number of BNNSs decreased from 15 to 4 with the mole ratio of boric acid and urea decreasing from 1:20 to 1:80, which was identified by SEM , TEM , AFM and BET measurements. More importantly, BNNSs presented an outstanding adsorption performance for estrone with the adsorption capacity of 249.15 mg E 1 /g BNNSs. The adsorption process could be best fitted by pseudo second-order kinetic model and the equilibrium data at different temperatures were well fitted by Langmuir isotherm model. The thermodynamics analysis revealed that E 1 adsorption on BNNSs was spontaneous (ΔG = −29.33 kJ mol −1 ), enthalpy-retarded (ΔH = 29.75 kJ mol −1 ), entropy-driven (ΔS = 198.26 J mol −1 K −1 ), and mostly chemical adsorption. The adsorption rates of E 1 in water were sharply enhanced with thinner BNNSs as absorbents and removal efficiency by BN-60 regenerated after 6 times was above 95%, it was shown that the surface areas, mesopores and remarkable structure played important roles in the adsorption process. The firmness of E 1 onto BNNSs and the stability of adsorption efficiency made BNNSs as a potential absorbent for efficient removal of E 1 from wastewater.

Identification of novel catabolic genes involved in 17β-estradiol degradation by Novosphingobium sp. ES2-1

Shunyao Li,Kai Sun,Xin Yan,Chao Lu,Michael Gatheru Waigi,Juan Liu,Wanting Ling

Summary Novosphingobium sp. ES2-1 is an efficient 17 β -estradiol (E2)-degrading bacterium, which can convert E2 to estrone (E1), then to 4-hydroxyestrone (4-OH-E1) for subsequent oxidative cracking. In this study, the molecular bases for this process were elucidated. Two novel monooxygenase systems EstP and EstO were shown to catalyse the oxygenation of E1 and 4-OH-E1, respectively. EstP was a three-component cytochrome P450 monooxygenase system consisting of EstP1 (P450 monooxygenase), EstP2 (ferredoxin) and EstP3 (ferredoxin reductase). Ultraperformance liquid chromatography-high resolution mass spectrometry (UPLC-HRMS) analysis revealed that EstP catalysed the 4-hydroxylation of E1 to produce 4-OH-E1. The resultant 4-OH-E1 was further oxidized by a two-component monooxygenase system EstO consisting of EstO1 (flavin-dependent monooxygenases) and EstO2 (flavin reductase). UPLC-HRMS combined with 1 H-nuclear magnetic resonance analysis demonstrated that EstO catalysed the breakage of C9-C10 to yield a ring B-cleavage product. In addition, the oxygenase component genes estP1 and estO1 exhibited contrary inductive behaviours when exposed to different steroids, suggesting that EstP1-mediated 4-hydroxylation was E2-specific, whereas EstO1-mediated monooxygenation might be involved in the degradation of testosterone, androstenedione, progesterone and pregnenolone. This also implied that the mechanisms of the catabolism of different steroids by the same microorganism might be partially interlinked.

The characteristics of oestrone mobility in water and soil by the addition of Ca-biochar and Fe–Mn-biochar derived from Litchi chinensis Sonn.

Tao Huan-yu,Ge Hui,Shi Jianghong,Liu Xiaowei,Guo Wei,Zhang Mengtao,Meng Yaobin,Li Xiao-yan

Abstract In this study, the effect of biochar (BC) derived from Litchi chinensis Sonn. and its modification, including Ca-biochar (Ca-BC) and Fe–Mn-biochar (Fe–Mn-BC), on the transportation of oestrone (E1) in water and soil was investigated. Fe–Mn-BC showed better adsorption ability than other types of biochar (BC, Ca-BC) under different conditions (humic acid, pH, ionic strength) in an aqueous environment. The maximum mass of sorbent at 298 K increased from 1.12 mg g −1 (BC) to 4.18 mg g −1 (Fe–Mn-BC). Humic acid had a greater impact on aqueous E1 adsorption on these biochars than did the pH and ionic strength. Fe–Mn-BC as a soil amendment had a great control of E1 transport in soil, and no leachate of E1 was observed in the column experiment. E1 mobility showed strong retardation in amended soil with Ca-BC ( R f  = 11.2) compared with raw soil ( R f  = 7.1). These results suggested that Fe–Mn-BC was more effective in controlling E1 transportation, and Fe–Mn-BC could be used as an alternative and inexpensive adsorbent to reduce E1 contaminants from water and soil. Construction of a microalgal-fungal spore co-culture system for the treatment of wastewater containing Zn(II) and estrone: Pollutant removal and microbial biochemical reactions

Xiaoman He,Dongyang Liu,Yue Teng,Hanzhi Wang,Qirui Wu,Wei Wang,Jingzheng Ren,Liandong Zhu

The co-culture system of Chlorella sorokiniana and Aspergillus oryzae has demonstrated exceptional tolerance and efficiency in the removal of pollutants from swine manure. This study evaluates the ability of the co-culture system to remove Zn(II) and estrone, while assessing the impact of these pollutants on the system's overall functionality. Results indicated that co-cultivation achieved higher biomass accumulation, peaking at 0.88 g/L after 96 h. Increasing estrone exposure concentration reduced photosynthetic activity and chlorophyll content, whereas Zn(II) exposure initially enhanced and later inhibited chlorophyll synthesis. Co-cultivation secreted extracellular polymeric substances, including protein-like and humus-like substances, to alleviate environmental stress and form algal-fungal community. After 96 h of cultivation, the removal efficiencies reached 86.44% for 1.5 mg/L Zn(II) and 84.55% for 20 mg/L estrone. The Quantitative Structure Activity Relationship model revealed a reduction in the ecotoxicity of estrone intermediate products to varying degrees. Metabolomics analysis showed that exposure to estrone and Zn(II) significantly boosted the production of Gibberellic acid, Indole-3-acetic acid, and Zeatin riboside in Chlorella sorokiniana , while reducing Abscisic Acid levels. Furthermore, the exposure led to an increase in various metabolites in the Tricarboxylic acid cycle of the co-cultivation system, influencing the synthesis and metabolism of key biochemical components like carbohydrates, lipids, and proteins. These findings elucidate the biochemical responses of Chlorella sorokiniana - Aspergillus oryzae co-culture system to pollutants and provide insights into its potential application in the treatment of wastewater containing endocrine disrupting chemicals and heavy metals.

Water depth alters the fate of estrone across the sediment–water interface in a typical inland lake

Linzhu Du,Shuhang Wang,Xia Jiang,Zhihao Wu,Arne Bratkic,Wei Guo

Water depth variation alters lake sediment environments, affecting the exchange of oxygen, trace elements, and nutrients at the sediment–water interface (SWI). However, there is still limited information on the in-situ fate of refractory emerging pollutants such as estrogenic compounds (ECs) at the SWI of different water depths in lakes. In this study, we integrated active sampling and diffusive gradients in thin film (DGT) technology to in-situ investigate the distribution, diffusion, and microbial responses of estrone (E1) at the SWI from the shallow-water zone (SZ), transition-water zone (TZ), and deep-water zone (DZ) in a typical inland lake of northern China. The surface sediment of the DZ accumulated most E1 (479.57 ± 194.90 ng/kg). In the vertical profile of the sediments, for SZ and TZ, E1 concentration in the sediment, porewater, and DGT fluctuated within a certain concentration range with increasing sediment depth. For the DZ, it exhibited a fluctuating and increasing trend with increasing depth. Dynamic migration analysis of E1 indicated partial resupply in each zone, and the resupply parameter R was 0.102 to 0.416 in the SZ, 0.111 to 0.384 in the TZ, and 0.084 to 0.374 in the DZ, respectively. The resupply capacity in the upper sediment layers (−1 to −4 cm) was higher for the TZ and DZ, whereas the SZ showed higher capacity in the lower layers (−5 to −9 cm). Based on the microbial sequencing results, Sulfuricurvum (typically present in microaerophilic and anoxic conditions) was predominant in the DZ (17.99 ± 15.4 %). Achromobacter (typical E1-degrading genus distributed in oxic environments) was more abundant in the SZ, which led to a higher E1 degradation potential in this zone. The results of this study shed light on the fate of E1 in the SWI of lakes at different water depths, facilitating a more precise control of estrogenic pollution in lake sediments.

Photocatalytic degradation of Estrone and Congo red by the magnetic antibacterial photocatalyst g-C3N4/CeO2/M-rGO under visible light and optimization by Box-Behnken statistical design (BBD)

Baogui Ye,Zhongwei Zhao,Hongmei Liu

Construction of multi-interface contact step-scheme photocatalyst is a promising pathway to achieve high-electron transfer efficiency for photodegradation Estrone and Congo red (CR). In this work, g-C 3 N 4 was selected as the main component, CeO 2 nanosheets with unique Ce 4+ →Ce 3+ conversion property and M-rGO were loaded on the surface of g-C 3 N 4 to construct magnetic photocatalyst, whose structure imparted reusability by magnetic retrieval and at the same time M-rGO acted as an efficient host for g-C 3 N 4 /CeO 2 composite photocatalyst. The photocatalytic performances were investigated with Estrone and CR, and the results showed that the composite photocatalyst g-C 3 N 4 /CeO 2 /M-rGO could degrade 71% of Estrone and 92% of CR. Free radical scavenger test showed that superoxide and hydroxyl radical species played a major role in the degradation. Furthermore, it was demonstrated that the magnetic photocatalyst g-C 3 N 4 /CeO 2 /M-rGO possessed strong antibacterial properties against Proteus mirabilis. A three-factor-three-level Box-Behnken design (BBD) was selected to optimize three greatly influential parameters: light irradiation time (min), the mass of photocatalyst (mg) and pollution concentration (ppm) by responding surface methodology. It was expected that the g-C 3 N 4 /CeO 2 /M-rGO can be used as a magnetic photocatalyst to achieve its valorization, which has very broad development prospects in the field of environmental remediation or catalysis.

Transformation of 17β-estradiol to estrone by unknown wild-type enzyme in commercial arylsulfatase from Helix pomatia

Jun Zhang,Ze-hua Liu,Ke-meng Zhao,Zhi Dang,Yun Liu,Yu Liu

This work for the first time reported the complete transformation of 17β-estradiol (E2) to estrone (E1) by unknown wild-type enzyme present in the widely used commercial arylsulfatase derived from Helix pomatia . It was found that acetate could effectively inhibit the unknown enzyme with a half inhibitory concentration (IC 50 ) of 140.9 μM, while phosphate and citrate showed no inhibition. Since the buffer solutions with phosphate and citrate have been used in the enzymatic hydrolysis of natural estrogen conjugates for decades, the transformation of E2 to E1 likely occurred during such procedure, inevitably leading to overestimated E1, but underestimated E2. It was further suggested that acetate should be used to prevent this undesirable transformation during the enzymatic hydrolysis of natural estrogen conjugates.

Bismuth impregnated biochar for efficient estrone degradation: The synergistic effect between biochar and Bi/Bi2O3 for a high photocatalytic performance

Ningyuan Zhu,Chunquan Li,Lingjun Bu,Cilai Tang,Sichu Wang,Pengfei Duan,Lunguang Yao,Jun Tang,Dionysios D. Dionysiou,Yonghong Wu

An innovative advanced oxidation process was successfully developed to photocatalytic-degradation of estrone through the synergistic effect of biochar and Bi/Bi 2 O 3 in bismuth-containing photocatalytic biochar (BiPB). The highest reaction rate constant ( k obs ) of estrone degradation by BiPB was 0.045 min −1 under the conditions of initial concentration of estrone =10.4 μmol L −1 , [BiPB] =1 g L −1 , pH = 7.0. The k obs was almost tenfold and more than 20 times than that of biochar without bismuth impregnation and pristine Bi/Bi 2 O 3 , respectively. The best photocatalytic performance of BiPB composites for the degradation of estrone was primarily attributed to generation of OH radicals. Impregnation of bismuth helped control the concentration of persistent free radicals (PFRs) and develop a hierarchical porous structure of biochar. The presence of biochar facilitated pre-concentration estrone on BiPB and improved the separation and transfer efficiency of charge carriers. The synergistic effect between biochar and Bi/Bi 2 O 3 contributed to the generation of OH radicals for estrone degradation under neutral pH conditions. The transformation pathway of estrone was also proposed based on the measured transformation products in the presence of BiPB. The high efficiency of BiPB composites indicated that this easily-obtained material was promising for estrone-wastewater treatment applications as a low-cost composite photocatalyst.

A Novel Estrone Degradation Gene Cluster and Catabolic Mechanism in Microbacterium oxydans ML-6

Lei Miao,Shanshan Sun,Tian Ma,Yousif Abdelrahman Yousif Abdellah,Yue Wang,Yaozu Mi,Haohao Yan,Guanjun Sun,Ning Hou,Xinyue Zhao,Chunyan Li,Hailian Zang

Global-scale estrone (E1) contamination of soil and aquatic environments results from the widespread use of animal manure as fertilizer, threatening both human health and environmental security. A detailed understanding of the degradation of E1 by microorganisms and the associated catabolic mechanism remains a key challenge for the bioremediation of E1-contaminated soil. Here, Microbacterium oxydans ML-6, isolated from estrogen-contaminated soil, was shown to efficiently degrade E1. A complete catabolic pathway for E1 was proposed via liquid chromatography-tandem mass spectrometry (LC-MS/MS), genome sequencing, transcriptomic analysis, and quantitative reverse transcription-PCR (qRT-PCR). In particular, a novel gene cluster (moc) associated with E1 catabolism was predicted. The combination of heterologous expression, gene knockout, and complementation experiments demonstrated that the 3-hydroxybenzoate 4-monooxygenase (MocA; a single-component flavoprotein monooxygenase) encoded by the mocA gene was responsible for the initial hydroxylation of E1. Furthermore, to demonstrate the detoxification of E1 by strain ML-6, phytotoxicity tests were performed. Overall, our findings provide new insight into the molecular mechanism underlying the diversity of E1 catabolism in microorganisms and suggest that M. oxydans ML-6 and its enzymes have potential applications in E1 bioremediation to reduce or eliminate E1-related environmental pollution.IMPORTANCE Steroidal estrogens (SEs) are mainly produced by animals, while bacteria are major consumers of SEs in the biosphere. However, the understanding of the gene clusters that participate in E1 degradation is still limited, and the enzymes involved in the biodegradation of E1 have not been well characterized. The present study reports that M. oxydans ML-6 has effective SE degradation capacity, which facilitates the development of strain ML-6 as a broad-spectrum biocatalyst for the production of certain desired compounds. A novel gene cluster (moc) associated with E1 catabolism was predicted. The 3-hydroxybenzoate 4-monooxygenase (MocA; a single-component flavoprotein monooxygenase) identified in the moc cluster was found to be necessary and specific for the initial hydroxylation of E1 to generate 4-OHE1, providing new insight into the biological role of flavoprotein monooxygenase.

Preliminary Study on the Removal of Steroidal Estrogens Using TiO2-Doped PVDF Ultrafiltration Membranes

Mingquan Wang,Fangshu Qu,Ruibao Jia,Shaohua Sun,Guibai Li,Heng Liang

Steroidal estrogens are a representative type of endocrine-disrupting chemical contaminant that has been detected in surface water. In this paper, modified polyvinylidene fluoride (PVDF) membranes were prepared by adding different amounts of polyvinyl pyrrolidone (PVP) and nano-TiO2particles. PVDF-PVP membrane adsorption, UV photolysis and PVDF-PVP-TiO2membrane photocatalysis performance were investigated by considering the rejection of estrone (E1) and 17β-estradiol (E2) in the cross-flow filtration experiments. The mechanism of photocatalytic degradation on TiO2-doped PVDF membranes was also evaluated. The results from the study indicated that adding PVP and nano-TiO2appropriately in PVDF membranes could be an effective method for better E1and E2 rejection due to adsorption and photocatalytic degradation. Keywords:steroidal estrogens;estrone;17β-estradiol;PVDF-PVP-TiO2ultrafiltration membrane;photocatalysis;water
