Graphical The dual-responsive polypyrrole-2,2'-Azobis(2-methylpropionamidine) dihydrochloride (AIPH)@Lauric acid nanoplatform integrates polypyrrole for photothermal therapy and AIPH for thermodynamic therapy, achieving synergistic colorectal cancer treatment. Under near-infrared irradiation, the system enables precise, oxygen-independent radical generation, enhancing tumor inhibition with minimal toxicity. This study highlights the platform's potential for minimally invasive cancer therapy. In this study, we introduce a novel nanoplatform, polypyrrole (PPy)-2,2′-Azobis[2-(2-imidazolin-2-yl)propane] dihydrochloride (AIPH)@lauric acid (LA) (PPy-AIPH@LA) nanoparticles (NPs), designed to overcome these limitations through synergistic photothermal therapy (PTT) and photodynamic therapy (PDT). This dual-responsive system incorporates PPy for efficient photothermal conversion, AIPH for thermos-responsive and oxygen-independent free radical generation, and LA as a thermally responsive encapsulation layer. The LA coating melts upon 808 nm near-infrared laser irradiation, releasing AIPH and free radicals to enable precise spatiotemporal activation of therapeutic effects. PPy-AIPH@LA demonstrates exceptional photothermal conversion efficiency (55.74%) and generates sufficient radicals to enhance PDT efficacy, even in hypoxic tumor microenvironments. In vitro studies revealed concentration-dependent tumor cell ablation and inhibition of migration, while in vivo experiments showed that the combined PTT-PDT treatment achieved an impressive 90.7% tumor growth inhibition rate in a mouse colon cancer cells CT-26 murine model, with no significant systemic toxicity. Molecular analyses further revealed modulations in pathways associated with tumor metabolism, apoptosis, and immune escape, highlighting the comprehensive therapeutic potential of this nanoplatform. These findings underscore the potential of PPy-AIPH@LA as a safe, effective, and minimally invasive nanotherapeutic platform for combating CRC and other solid tumors.

Microwave ablation (MWA) is a novel treatment modality that can lead to the death of tumor cells by heating the ions and polar molecules in the tissue through high-speed vibration and friction. However, the single hyperthermia is not sufficient to completely inhibit tumor growth. Herein, a thermodynamic cancer-therapeutic modality has been fabricated which could be able to overcome hypoxia's limitations in the tumor microenvironment. Using thermo-sensitive liposomes (TSLs) and oxygen-independent radical generators (2,2’-azobis[2-(2-imidazolin-2-yl)propane]dihydrochloride [AIPH]), a nano-drug delivery system denoted as ATSL is developed for efficient sequential cancer treatment. Under the microwave field, the temperature rise of local tissue could not only lead to the damage of tumor cells but also induce the release of AIPH encapsulated in ATSL to produce free radicals, eliciting tumor cell death. In addition, the ATSL developed here would avoid the side effects caused by the uncontrolled diffusion of AIPH to normal tissues. The ATSLs have shown excellent therapeutic effects both in vitro and in vivo, suggesting its highly promising potential for clinic.

In this study, we developed an innovative Cu 2-x Se/PDA/AIPH nanoparticle platform that combines photothermal therapy and chemotherapy for effective tumor treatment. The Cu 2-x Se nanoparticles, known for their strong near-infrared (NIR) absorption, were encapsulated within a polydopamine (PDA) and 2,2′-azobis[2-(2-imidazolin-2-yl)propane] dihydrochloride (AIPH) matrix. Upon NIR irradiation, the platform triggers localized heating and subsequent thermal decomposition of AIPH, releasing ROS to induce significant oxidative damage in tumor cells. In vitro and in vivo experiments demonstrated that Cu 2-x Se/PDA/AIPH nanoparticles exhibit excellent biocompatibility, effective photothermal conversion, and potent anticancer efficacy. This multifunctional nanosystem offers a promising approach for enhancing tumor therapy by combining PTT with ROS-mediated chemotherapy.

Drug-resistant bacterial infections pose a significant threat to global public health. Furthermore, the formation of biofilms makes traditional antibiotic treatments significantly less effective in killing multidrug-resistant (MDR) bacteria. In this work, we prepared a molybdenum disulfide (MoS2) nanosphere-based nanocomposite that utilized both photothermal therapy (PTT) and chemodynamic therapy (CDT) for effectively eradicating biofilms. A layer of manganese dioxide (MnO2) was adsorbed onto the surface of the MoS2 nanospheres by redox and electrostatic adsorption methods, and the resulting MoS2/MnO2 were loaded with 2,2′-azobis[2-(2-imidazolin-2-yl)propane]-dihydrochloride (AIPH) to form the MoS2/MnO2-AIPH nanocomposite (MMA). The microenvironment of biofilms is slightly acidic, lacks oxygen, and has a high concentration of glutathione (GSH). The MnO2 was capable of reacting with GSH to deplete it while also generating •OH radicals for CDT. In the presence of NIR light (808 nm), the temperature increase brought about by PTT further enhanced the CDT efficacy synergistically. The results showed that the photothermal conversion efficiency of the MMA was 40.5% at a concentration of 100 μg/mL, and the biofilm eradication rate of both MDR Escherichia coli and MDR Staphylococcus aureus was above 90%. The high bacterial inhibition rate (above 96%), as well as the excellent biosafety and biocompatibility of the MMA nanocomposite enabled it to effectively promote wound healing, which has significant implications in treating bacterial infections and promoting wound healing in clinical settings.

This research introduces FAM (FeS2-AIPH@Membrane), a nanoparticle system that synergizes photothermal therapy (PTT), chemodynamic therapy (CDT), and thermodynamic therapy (TDT) for targeted cancer treatment. FAM utilizes FeS2 for photothermal conversion and AIPH for radical generation, encapsulated within a tumor cell membrane to enhance targeting and immune evasion. The synthesis of FeS2 with AIPH was optimized, yielding nanoparticles with excellent stability and photothermal properties. Designed to release drugs in acidic tumor environments, FAM increases cytotoxicity at the target site. Under NIR irradiation in vitro, FAM nanoparticles mediated tumor cell death through concurrent reactive oxygen species production and localized hyperthermia. Cellular uptake studies confirmed enhanced intracellular concentrations due to homologous targeting. In vivo tests on MNNG/HOS tumor-bearing mice showed that FAM significantly reduced tumor volumes without systemic toxicity. These results demonstrate FAM’s potential as a safe, effective cancer theranostics platform, integrating PTT, CDT, and TDT into a single treatment modality, offering a more targeted, efficient, and less invasive cancer treatment alternative.

Tumors have become the biggest obstacle to human health, and there are various treatment methods at present. Photothermal therapy (PTT) is usually ineffective and does not inhibit tumor progression due to the inability of the lasers to penetrate deeply. Therefore, most existing studies chose a 1064 nm laser with stronger penetrating power; meanwhile, studies have shown that the inclusion of harmful free radicals can significantly improve the antitumor efficacy. Herein, TiO nanosheets (NSs) were creatively prepared and encapsulated with an alkyl radical generator {2,2′-azobis[2-(2-imidazoline-2-yl)propane] dihydrochloride, [AIPH]} in sodium alginate (ALG) hydrogel for effective tumor killing by PTT and pairing with dangerous free radicals. TiO NSs were obtained by the liquid-phase exfoliation method, together with AIPH, which were in situ coencapsulated multifunctional hydrogels formed by the combination of Ca2+ and ALG. This ALG hydrogel could enrich TiO NSs and AIPH at the tumor site for a long time, and through the excellent photothermal properties of TiO NSs, AIPH could slowly and effectively generate alkyl radicals at the tumor site, which, in turn, gave it a better antitumor effect compared with that of TiO NSs in the deep hypoxic environment of the tumor. The AIPH + TiO + ALG hydrogel has distinctive anticancer capabilities based on the results of both in vivo and in vitro experiments. This material also has good biosafety. By combining PTT and free radical treatment, this work provides a novel therapeutic method to achieve oxygen-independent free radical production and enhance therapeutic efficacy.

Drug-resistant bacterial infection is a serious threat to the survival and health of human. Clinically, the conventional therapy for subcutaneous infections generally involves the systemic administration of high dose of antibiotics and frequently causes detrimental side effects in normal tissues. To overcome this issue, an acidity-responsive near infrared (NIR) light-triggered injectable hydrogel was designed for non-antibiotic local treatment. Herein, poly(ethylene glycol) diacrylate (PEGDA) was used as hydrogel matrix, polyoxometalate (POM) as acidity-enhanced photothermal agent and 2,2′-azobis[2-(2-imidazolin-2-yl) propane] dihydro-chloride (AIPH) as thermo-responsive initiator. After the hydrogel precursor solution (AIPH/POM/PEGDA) was injected into the acidic infected tissue, the aggregated POM with strong photothermal property was restricted to the site of abscess. Upon excitation at 1060 nm laser, the heat generated from POM would trigger alkyl radical formation from AIPH and thereafter achieving NIR-triggered in situ gel. Consequently, the POM and AIPH were trapped in Gel(AIPH/POM) and exhibited long-term retention in the abscess for further repeated local photothermal/thermodynamic antibacterial therapy. In vitro antibacterial evaluation indicated that the system could effectively eradicate drug-resistant bacteria through destroying bacteria integrity and producing the noxious alkyl radical under NIR light illumination. Moreover, the bacterial burden of abscess was significantly relieved and more collagens were deposited after the administration of Gel(AIPH/POM) and repeated NIR light. Meanwhile, no apparent adverse effect was observed during the treatment. This work not only provides a light-triggered gelation for local photothermal/thermodynamic synergistic therapy, but also offers a novel strategy for the rational design of light-responsive intelligent antibacterial hydrogels.

Due to the rapid proliferation of tumor cells leading to high oxygen consumption, solid tumors generally have the characteristics of hypoxia, which greatly limits the effect of photodynamic therapy sensitive to oxygen levels. Here, we report the construction of a photodynamic/photothermal synergistic theranostic agent, which mainly utilizes oxygen-independent free radicals generated from a thermally decomposable free radical initiator and in situ heating to kill cancer cells, overcoming the poor tumor therapeutic effect in a hypoxic environment. In this case, the 2,2′-azobis-[2-(2-imidazolin-2-yl)propane] dihydrochloride (AIPH) molecule, as a thermally decomposable free radical initiator, is loaded on polydopamine-coated PdAu alloy nanoparticles via π–π stacking (named as AIPH-PdAu@PDA). A self-assembly strategy was applied by using gadolinium-bovine serum albumin (Gd-BSA) complexes to encapsulate the AIPH-PdAu@PDA nanoparticles, resulting in the final magnetic resonance/computed tomography/photoacoustic triple-modal imaging guided and single laser (808 nm) activated photodynamic/photothermal therapy theranostic agent AIPH-PdAu@PDA@Gd-BSA, denoted as APPG. Systematic evaluations in vitro demonstrate that this interesting theranostic platform can rapidly generate high heat under irradiation from the 808 nm laser, thereby inducing the initiator AIPH molecule to produce oxygen-independent free radicals and displaying excellent triple-modal imaging performance. Therefore, the proposed triple-modal imaging-guided single laser activated synergistic therapy nanocomposite is a promising platform for imaging-guided cancer treatment.

Ferroptosis has received ever-increasing attention due to its unparalleled mechanism in eliminating resistant tumor cells. Nevertheless, the accumulation of toxic lipid peroxides (LPOs) at the tumor site is limited by the level of lipid oxidation. Herein, by leveraging versatile sodium alginate (ALG) hydrogel, a localized ferroptosis trigger consisting of gambogic acid (GA), 2,2′-azobis [2-(2-imidazolin-2-yl) propane] dihydrochloride (AIPH), and Ink (a photothermal agent), was constructed via simple intratumor injection. Upon 1064 nm laser irradiation, the stored AIPH rapidly decomposed into alkyl radicals (R•), which aggravated LPOs in tumor cells. Meanwhile, GA could inhibit heat shock protein 90 (HSP90) to reduce the heat resistance of tumor cells, and forcefully consume glutathione (GSH) to weaken the antioxidant capacity of cells. Systematic in vitro and in vivo experiments have demonstrated that synchronous consumption of GSH and increased reactive oxygen species (ROS) facilitated reduced expression of glutathione peroxidase 4 (GPX4), which further contributed to disruption of intracellular redox homeostasis and ultimately boosted ferroptosis. This all-in-one strategy has a highly effective tumor suppression effect by depleting and generating fatal active compounds at tumor sites, which would pave a new route for the controllable, accurate, and coordinated tumor treatments.

Aim: This study aimed to develop a sonodynamic–chemodynamic nanoparticle functioning on glutathione depletion in tumor immunotherapy. Materials & methods: The liposome-encapsulated 2,2-azobis[2-(2-imidazolin-2-yl) propane] dihydrochloride (AIPH) and copper–cysteine nanoparticles, AIPH/Cu-Cys@Lipo, were synthesized with a one-pot method. 4T1 cells were injected into female BALB/c mice for modeling. Results: AIPH/Cu-Cys@Lipo was well synthesized. It generated alkyl radicals upon ultrasound stimulation. AIPH/Cu-Cys@Lipo promoted the generation of -OH via a Fenton-like reaction. Both in vitro and in vivo experiments verified that AIPH/Cu-Cys@Lipo significantly inhibited tumor development by decreasing mitochondrial membrane potential, activating CD4+ and CD8+ T cells and promoting the expression of IL-2 and TNF-α. Conclusion: AIPH/Cu-Cys@Lipo provides high-quality strategies for safe and effective tumor immunotherapy.

The efficacy of sonodynamic therapy (SDT) is largely dependent upon oxygen availability to generate deleterious reactive oxygen species, and as such, hypoxic microenvironments greatly constrain the efficacy of SDT. Development of free radical generators that are not dependent on oxygen and related combination treatment strategies thus have the potential to enhance the antitumor potential of SDT. Combined treatment strategies are expected to improve the efficacy of sonodynamic antitumor therapy. As metal–organic framework (MOF) platforms are highly amenable to integration with other therapeutic approaches, we herein report the development of tumor microenvironment (TME)-responsive nanoparticles constructed by embedding the azo initiator 2,2′-azobis[2-(2-imidazolin-2-yl)propane]dihydrochloride (AIPH) into hypoxia-triggered copper metal–organic framework (Cu-MOF) nanovectors to achieve synergistic sono-chemodynamic therapy in an orthotopic murine pancreatic carcinoma model system. When exposed to hypoxic conditions within the TME, this Cu-MOF structure underwent degradation, leading to the release of Cu2+ and AIPH. Cu2+ was then able to deplete local glutathione stores, resulting in the reduction of Cu2+ to Cu+, which then reacts with endogenous H2O2 in a Fenton-like reaction to yield cytotoxic hydroxyl radicals (•OH) for chemodynamic therapy. When exposed to ultrasound irradiation, AIPH further degraded in an oxygen-independent manner to yield nitrogen bubbles and alkyl radicals, the former of which enhanced the ability of these nanoparticles to penetrate deeply into the tumor. The resultant radicals induced substantial DNA damage and apoptotic cell death within target tumors under different levels of oxygen availability. As such, this hypoxic TME-responsive synergistic sono-chemodynamic approach offers an ideal means of achieving oxygen-independent free radical generation and enhanced treatment efficacy.
