概述

结晶紫是一种可与细胞核中DNA结合的碱性染料，主要用于细胞核染色；甲基紫染色切片需采用水溶性封固剂封固，因局部脱水作用会破坏红色多染性。结晶紫甲醇溶液 (2.5%) 由结晶紫、甲醇等组成，是一种组织或细胞染色时常用的可以把细胞核染成深紫色的染色液，一般和其他染色液配合使用或稀释后使用。

操作步骤（仅供参考）

1. 样品处理

石蜡切片：先在二甲苯中脱蜡5-10分钟，更换新鲜二甲苯后再次脱蜡5-10分钟；随后依次放入无水乙醇5分钟、90%乙醇2分钟、70%乙醇2分钟，最后用蒸馏水浸泡2分钟。
冰冻切片：直接用蒸馏水浸泡2分钟即可。
培养细胞：先用4%多聚甲醛固定10分钟以上，蒸馏水洗涤2分钟，更换新鲜蒸馏水后再次洗涤2分钟。

2. 结晶紫染色

将上述处理好的样品，放入结晶紫染色液中染色（染色时间可根据染色结果及实际要求灵活调整）；染色完成后，用蒸馏水或自来水充分洗涤，即可进行观察和拍照。

注意事项

1. 一般稀释 20 倍使用。

2. 首次使用本试剂时，建议先取1～2个样品进行预实验，确认染色效果及适配性。

3. 结晶紫对人体有轻微危害，操作时请做好适当防护。

4. 为保障您的安全和健康，请规范操作本试剂。

5. 本产品仅用于科研用途，严禁用作其他用途。

Crystal violet is a basic dye that binds to DNA in the cell nucleus and is primarily used for nuclear staining. Methyl violet-stained sections should be mounted using a water-soluble mounting medium, as local dehydration can disrupt its polychromatic red staining properties.

Crystal Violet Methanol Solution (2.5%) is composed of crystal violet and methanol. It is a commonly used staining solution in histology or cytology that stains the nucleus a deep purple. It is typically used in combination with other staining solutions or after dilution.

Procedure (For Reference Only)

1. Sample Preparation

Paraffin Sections:

First, dewax in xylene for 5-10 minutes, then replace with fresh xylene and dewax again for 5-10 minutes. Subsequently, sequentially transfer the sections into absolute ethanol for 5 minutes, 90% ethanol for 2 minutes, 70% ethanol for 2 minutes, and finally soak in distilled water for 2 minutes.

Frozen Sections:

Soak directly in distilled water for 2 minutes.

Cultured Cells:

Fix with 4% paraformaldehyde for at least 10 minutes, then wash with distilled water for 2 minutes. Replace with fresh distilled water and wash again for 2 minutes.

2. Crystal Violet Staining

Place the prepared samples into the crystal violet staining solution for staining (the staining time can be adjusted flexibly based on staining results and practical requirements). After staining, rinse thoroughly with distilled water or tap water, then proceed to observation and photography.

Notes

1. It is generally recommended to dilute 20-fold before use.

2. When using this reagent for the first time, it is recommended to conduct a pilot experiment with 1–2 samples to confirm the staining effect and compatibility.

3. Crystal violet is slightly hazardous to humans. Please take appropriate protective measures during operation.

4. To ensure your safety and health, please handle this reagent according to standard operating procedures.

5. This product is intended for research use only and is strictly prohibited for any other purposes.
