概述
结晶紫（Crystal Violet）又称甲紫，属于碱性染料，能溶于水、乙醇，可以把组织和细胞核染成紫色。结晶紫主要应用于细胞学、组织学和细菌学等方面，是一种优良的染色剂。
使用方法
一、样品处理
1. 对于石蜡切片
(1) 二甲苯中脱蜡5~10 min。
(2) 换用新鲜的二甲苯，再脱蜡5~10 min。无水乙醇5 min。
(3) 90% 乙醇2 min。70% 乙醇2 min。蒸馏水2 min。
2. 对于冰冻切片
取出恢复至室温后蒸馏水2 min。
3. 对于培养细胞
(1) 用4% 多聚甲醛固定10 min以上（或采用如下步骤：每25cm²加入PBS/甲醇溶液 5 mL，静置2 min，弃液；加入新鲜甲醇5 mL，静置10 min，弃液）。
(2) 蒸馏水洗涤2 min。
(3) 更换新鲜的蒸馏水，再洗涤2 min。
二、结晶紫染色
1. 结晶紫水溶液 (1%) 染色10 min（可以根据染色结果和要求调整时间）。
2. 用蒸馏水或自来水充分洗涤，进行观察和拍照。

Crystal Violet, also known as Gentian Violet, is a basic dye soluble in water and ethanol, which stains tissues and cell nuclei purple. It is an excellent staining agent mainly used in cytology, histology, bacteriology, and other fields.
Protocol
I. Sample Preparation
1. For Paraffin Sections
(1) Deparaffinize in xylene for 5–10 min.
(2) Replace with fresh xylene and deparaffinize for another 5–10 min; treat with absolute ethanol for 5 min.
(3) 90% ethanol for 2 min; 70% ethanol for 2 min; distilled water for 2 min.
2. For Frozen Sections
Allow sections to equilibrate to room temperature, then rinse with distilled water for 2 min.
3. For Cultured Cells
(1) Fix with 4% paraformaldehyde for ≥10 min (or follow this alternative: add 5 mL of PBS/methanol solution per 25 cm², incubate for 2 min, discard the solution; add 5 mL of fresh methanol, incubate for 10 min, discard the solution).
(2) Rinse with distilled water for 2 min.(3) Replace with fresh distilled water and rinse for another 2 min.
II. Crystal Violet Staining
Stain with 1% Crystal Violet Aqueous Solution for 10 min (adjust time according to staining results and requirements).
Wash thoroughly with distilled or tap water, then observe and photograph.

Precautions
Crystal Violet is harmful to humans; please take appropriate protective measures.
It is recommended to perform a pilot experiment on 1–2 samples when using this reagent for the first time.
