Glutaminase (EC 3.5.1.1) is an amidohydrolase that catalyzes the hydrolysis of glutamine to L-glutamate and ammonia. It plays a crucial regulatory role in nitrogen metabolism, particularly in modulating free ammonia levels and urea metabolism. Assay Principle: GLS catalyzes the hydrolysis of glutamine to L-glutamate and ammonia. The activity of GLS is calculated by measuring the rate of ammonia increase using Nessler's reagent.
