It is mainly used for the isolation of human peripheral blood lymphocytes
The entire process, including samples, reagents and experimental environment, must be carried out under the condition of 20±2℃ (reagents need to be rewarmed to 20℃ in summer and 23℃ in winter).
Matters needing attention:
1.Discussion on the effect of blood separation medium
A. Umbilical cord blood: The interval between umbilical cord blood collection and cell separation is generally more than 2 hours, and refrigerated storage and transportation are inevitable.
B. Special case: For samples from subjects who have used immunomobilizer drugs, ordinary density separation medium has poor separation effect with a large amount of granulocyte contamination, and thus is not applicable.
2.It is not recommended to use high-polymer plastic products (such as polystyrene) in this experiment. Non-electrostatic/low-electrostatic centrifuge tubes and un-alkali-treated glass products should be used instead. Electrostatic interaction will cause cells to adhere to the wall, and alkali treatment will turn the glass surface into a rough surface, both of which will affect the cell separation effect.
3.Suctioning too much lymphocyte layer and separation medium layer will lead to sucking out granulocytes at the junction of the separation medium, thus increasing the number of contaminated granulocytes in the product.
4.Suctioning too much supernatant above the lymphocyte layer will lead to contamination of plasma protein and platelets.
5.Improper dilution method will reduce cell yield and viability. Dilution requirement: use buffer or medium free of calcium and magnesium ions. If the blood sample is diluted, the centrifugal force and centrifugation time need to be appropriately reduced during the separation process.
