Induction of heme oxygenase-1 by hemin alleviates acute lung injury through inhibition of NLRP3 inflammasome assembly

Gao Xinyue,You Long,Qi Luyao,Jin Lei,Zhang Jianmei,Liu Yanan,Niu Zhiqiang

As a disease of high mortality rate, acute lung injury (ALI) has been getting increased attention. Heme oxygenase-1 (HO-1), which plays an essential role in alleviating the pathological symptoms of ALI. There have few studies noted that the improvement effect of HO-1 on ALI may associated to NOD-like receptor protein 3 (NLRP3) inflammasome. However, the roles of HO-1 in the process of NLRP3 inflammasome activation is remains unknown. This study investigated the mechanism that HO-1 blocking NLRP3 inflammasome in ALI. The sepsis model induced by LPS was used in C57BL6 mice, LPS triggered obviously pulmonary damage and edema formation, increased the expression of NLRP3 inflammasome component proteins and IL-1β, but those negative effects were reversed by the pretreatment of hemin. NOD-like receptor signaling pathway was down-regulated after hemin treatment through RNA-Seq. Meanwhile, hemin pretreatment exerted significantly inhibit effect on the activation of NLRP3 inflammasome, and the component proteins of NLRP3 inflammasome were not influenced in immortalization of bone marrow-derived macrophages (iBMDMs). Moreover, the immunoprecipitation and immunofluorescence experiments were indicated that HO-1 could bind to NLRP3, silencing HO-1 prevented the block effect of hemin on NLRP3 inflammasome activation. Our study suggested that hemin-inhibited NLRP3 inflammasome activation involved HO-1 bind to NLRP3 and alleviated ALI mice induced by LPS.

Silver-decorated lysozyme-hemin nanocomposite loaded into thermosensitive hydrogel for on-demand management of MRSA-infected wounds

Guannan Le,Xiaoyu Ma,Ling Cai,Wei Wei,Jinhuan Li,Jin Chen

The effectiveness of antibiotic treatment for bacterial-infected wounds has been greatly hampered by the increasing antimicrobial resistance (AMR) caused by improper antibiotic use. It is thereby demanding for the deployment of antibiotic-free and on-demand therapeutic strategies, such as chemodynamic therapy (CDT) and photothermal therapy (PTT). Here, via a desolvation method, L ysozyme- H emin nanocomposite was firstly prepared and further decorated by S ilver nanoparticles to form LHS NPs, which was then incorporated into a thermosensitive hydrogel. The LHS NPs demonstrated remarkable peroxidase-like activity and photothermal effect, with a specific activity of 2.59 U/mg and a photothermal conversion efficiency of 53.89 %. Consequently, it was found that the formed LHS NPs exhibited superior antibacterial activity against both Gram-negative and Gram-positive bacteria without the using of antibiotics. Moreover, through the optimization of nanozymes and hydrogels, the obtained nanozyme-hydrogel composite could adhere to wound sites of various shapes within 2 min upon NIR irradiation, enabling the on-demand wound treatment. Notably, in a methicillin-resistant Staphylococcus aureus ( MRSA )-infected wound model, the nanocomposite demonstrated excellent antibacterial activity and greatly sped up wound healing. Therefore, the LHS nanoparticles-loaded thermosensitive hydrogel as an integrated CDT/PTT therapeutic nanoplatform holds enormous potential in combating drug-resistant bacteria towards the complex infected wounds management.

Solvent-free grinding synthesis of Hemin@P2Mo18 with peroxidase-mimicking activity for dopamine detection in bovine serum

Chu Shiqi,Yang Lian,Chi Zhongmei,Ma Jiahui,Wang Huanxiang,Yan Lihe,Li Zhaoye,Liu Guocheng

Dopamine (DA) in bovine serum served as a vital physiological biomarker for evaluating livestock health status and conducting translational biomedical research. Accurate and rapid detection of DA was therefore essential for agricultural breeding monitoring and clinical biological analysis. In this work, a novel Hemin@P 2 Mo 18 nanocomposite with peroxidase-mimicking activity was successfully prepared via a facile, green solvent-free grinding strategy. The introduction of water-soluble H 6 P 2 Mo 18 effectively suppressed the aggregation of free hemin molecules and yielded solid-phase Hemin@P 2 Mo 18 . Relative to natural peroxidases, the synthesized Hemin@P 2 Mo 18 featured convenient recovery and excellent reusability. Given the excellent peroxidase-mimicking activity of Hemin@P 2 Mo 18 , a sensitive and selective colorimetric platform was constructed for DA detection. Moreover, the proposed approach showed high selectivity against common interfering substances and excellent stability under different conditions. It was successfully applied to the quantitative determination of DA in bovine serum samples with satisfactory recovery rates ranging from 96.8% to 100.2%. This study provided a green and efficient solvent-free grinding synthesis strategy for high-performance POM-based nanozymes. It also offered a feasible and practical approach for rapid biomarker detection in complex biological matrices, which showed great application potential in veterinary monitoring and biosensing analysis. A novel dual signal and label-free electrochemical aptasensor for mucin 1 based on hemin/graphene@PdPtNPs

Guojuan Zhang,Zhiguang Liu,Lifang Fan,Yujie Han,Yujing Guo

A dual signal and label-free electrochemical aptasensor for mucin 1 was constructed based on hemin/ [email protected] nanocomposite ( [email protected] ). Hemin attached on the graphene surface not only improves the solubility of graphene and acts as an in-situ electrochemical probe but also exhibits excellent peroxidase-like properties to electrocatalyze the reduction of H 2 O 2 . PdPtNPs also show outstanding catalytic capacity to the reduction of H 2 O 2 and provide numerous binding sites for loading dDNA (mucin 1 aptamer and cDNA) to form the sensing interface. In the presence of mucin 1, due to the specific affinity between aptamer and mucin 1, double helix would be induced dissociation and the aptamer would be pulled off from the electrode. As a result, the electrochemical signals of hemin and H 2 O 2 were recovered. Based on these properties, the label-free and sensitive dual signal electrochemical biosensor for mucin 1 detection has been developed. The one is differential pulse voltammetry (DPV) signal of hemin and the other is chronoamperometry signal arisen from the catalytic reduction of H 2 O 2 . The linear ranges for mucin 1 were 8.0 pg mL −1 to 80 ng mL −1 and 0.8 pg mL −1 to 80 ng mL −1 with the limit of detection 2.5 pg mL −1 and 0.25 pg mL −1 by DPV and chronoamperometry, respectively. The recovery of mucin 1 in human blood serum samples was from 95.0% to 104.2%. The detection platform does not need signal labeling which greatly reduced the sophisticated and expensive procedures. The aptasensor provide a promising strategy for the determination of mucin 1 in clinical diagnostics.

Fabrication of polydopamine/hemin-cyclodextrin supramolecular assemblies for mimicking natural peroxidases and their sensitive detection of cholesterol

Hongchao Ma,Yingying He,Lulu Xu,Yanhui Wei

The development of peroxidase mimics can overcome shortcomings of natural peroxidases and expand their biological applications. Herein, inspired by Hemeproteins family such as peroxidase and cytochromes , polydopamine/hemin-β-cyclodextrin (PDA/Hemin-CD) supramolecular assemblies were prepared to mimick natural peroxidases using hemin as the active center. Polydopamine acts as skeleton material which could encapsulate hemin molecules during the self-assembly procedure. Mono-(6-Mercapto-6-deoxy)-β-Cyclodextrin (β-CD-SH) were decorated on the surface of PDA/Hemin serving as the molecular recognition elements. Interestingly, the addition of cholesterol could “turn off” the peroxidase activity of PDA/Hemin-CD due to the specific recognition of β-CD to cholesterol. The excellent peroxidase activity as well as molecular recognition nature of PDA/Hemin-CD make it promising candidate to construct biosensors . Based on this, a colorimetric assay for the detection of cholesterol was developed.

Cu–hemin metal-organic frameworks with peroxidase-like activity as peroxidase mimics for colorimetric sensing of glucose

Liu Fenfen,He Juan,Zeng Mulang,Hao Juan,Guo Qiaohui,Song Yonghai,Wang Li

Abstract In this work, a facile strategy to synthesize Cu–hemin metal-organic frameworks (MOFs) with peroxidase-like activity was reported. The prepared Cu–hemin MOFs were characterized by various techniques such as scanning electron microscopy, transmission electron microscopy, X-ray powder diffraction, Fourier transform infrared spectroscopy, UV–visible absorbance spectra, and so on. The results showed that the prepared Cu–hemin MOFs looked like a ball-flower with an average diameter of 10 μm and provided a large specific surface area. The Cu–hemin MOFs possessing peroxidase-like activity could be used to catalyze the peroxidase substrate of 3,3,5,5-tetramethylbenzidine in the presence of H 2 O 2 , which was employed to detect H 2 O 2 quantitatively with the linear range from 1.0 μM to 1.0 mM and the detection limit was 0.42 μM. Furthermore, with the additional help of glucose oxidase, a sensitive and selective method to detect glucose was developed by using the Cu–hemin MOFs as catalyst and the linear range was from 10.0 μM to 3.0 mM and the detection limit was 6.9 μM. This work informs researchers of the advantages of MOFs for preparing biomimetic catalysts and extends the functionality of MOFs for biosensor application. Graphical Abstract

Colorimetric Detection of Copper(II) Ion Using Click Chemistry and Hemin/G-Quadruplex Horseradish Peroxidase-Mimicking DNAzyme

Chenchen Ge,Quan Luo,Dou Wang,Shiming Zhao,Xiaoling Liang,Luxin Yu,Xuerong Xing,Lingwen Zeng

G-quadruplex-forming sequence can be formed through a copper(I) ion (Cu+)-catalyzed click chemistry between azide- and alkyne-modified short G-rich sequences in aqueous solution, eliminating immobilization and washing steps of conventional assays. The source for Cu+ was generated from the reduction of Cu2+ with the reductant of sodium ascorbate. In the presence of hemin and K+, the self-assembly of hemin/G-quadruplex structure has the activity of horseradish peroxidase (HRP), which can catalyze its colorless substrate tetrazmethyl benzidine (TMB) into a colored product. Hence, the concentration of Cu2+ can be evaluated visually for qualitative analysis according to the color change of the solution, and the optical density (OD) value of the resulting solution at 450 nm was also recorded using a microplate reader for quantitative analysis.

Polypyrrole-hemin-reduce graphene oxide: rapid synthesis and enhanced electrocatalytic activity towards the reduction of hydrogen peroxide

Wenjing Huang,Qingli Hao,Wu Lei,Lihua Wu,Xifeng Xia

An efficient and eco-friendly microwave-assistant method is developed to synthesize a ternary composite of polypyrrole-hemin-reduced graphene oxide (PPY-He-RGO). The polymerization of the pyrrole monomer and the reduction of graphene oxide are performed simply by microwave heating without using a strong reducing or oxidizing agent in an isopropanol/H2O mixed medium. Hemin molecules are immobilized on reduced graphene oxide (RGO) sheets and can still retain high electrocatalytic activity toward the reduction of H2O2 in the final composite. The conducting RGO and polypyrrole with a well-controlled nanostructure provide a highly conductive network to the ternary composite, which can promote the electron transfer between hemin, analytes and electrodes, leading to an improved electrocatalytic activity. The PPY-He-RGO can act as a third-generation mediator and mimic enzyme for the fabrication of a hydrogen peroxide biosensor. The as-prepared PPY-He-RGO electrode exhibits a high sensitivity to H2O2 with a low detection limit of 0.13 μm. The efficient microwave heating provides an opportunity for large-scale production of PPY-He-RGO ternary nanocomposites as a kind of mimic enzyme for biosensors.

Cu-Hemin Metal-Organic-Frameworks/Chitosan-Reduced Graphene Oxide Nanocomposites with Peroxidase-Like Bioactivity for Electrochemical Sensing

Li Wang,Han Yang,Juan He,Yayun Zhang,Jie Yu,Yonghai Song

Herein, a Cu-hemin metal-organic-frameworks (MOFs)/chitosan (CS)-reduced graphene oxide (CS-rGO) nanocomposite with unique peroxidase-like bioactivity and good electrical conductivity was prepared for electrochemical H 2 O 2 sensing for the first time. The prepared Cu-hemin MOFs/CS-rGO nanocomposites were well characterized by scanning electron microscopy, energy dispersive X-ray spectroscopy, thermogravimetric analysis, Fourier transform infrared spectroscopy, X-ray powder diffraction, UV–vis spectroscopy and electrochemical techniques. The results showed that after the Cu-hemin MOFs were formed on the CS-rGO surface, the crystalline structure of the Cu-hemin MOFs was kept while the size of Cu-hemin MOFs was decreased and the electrical conductivity of the nanocomposites was enhanced greatly as compared with that of Cu-hemin MOFs. The unique peroxidase-like bioactivity and good electrical conductivity as well as some novel properties of Cu-hemin MOFs/CS-rGO nanocomposites resulted in perfect electrochemical performances towards the reduction of hydrogen peroxide (H 2 O 2 ), superior to some mimic enzymes. The Cu-hemin MOF/CS-rGO was used to construct a novel H 2 O 2 electrochemical sensor which exhibited wide linear range (0.065–410   μM) and low detection limit (0.019   μM). The study might provide a new proposal for preparing perfect hemin-based materials as mimetic peroxidase in the electrochemical sensing.

Effect of annealing temperature and element composition of titanium dioxide/graphene/hemin catalysts for oxygen reduction reaction

Yuewu Zhao,Linqun Zhang,Wei Wei,Ying Li,Anran Liu,Yuanjian Zhang,Songqin Liu

The oxygen reduction reaction (ORR) plays an important role at the cathode of fuel cells in practical applications. Herein, a titanium dioxide/graphene supported hemin (TiO2/Gr/Hem) composite material with a flower-like superstructure was successfully prepared through a two-step solvothermal reaction. By a further heat-treatment at 300–900 °C, the electrocatalytic activity of the as-obtained catalysts was examined, and it was found that the pyrolysis at 700 °C gave rise to the best catalytic activity for the ORR in alkaline media. This heat-treatment temperature was found to be crucial in determining the activity and stability of catalysts, due to the enhanced structural defects, active sites, geometrical complexity, and larger fraction of the pyridinic nitrogen and pyrrolic nitrogen groups. The titanium dioxide/graphene (TiO2/Gr) and graphene/hemin (Gr/Hem) were also studied and compared, and it was revealed that the catalytic activity of TiO2/Gr/Hem catalysts for ORR can be further enhanced. In addition, the chemically bonded element iron in the heat-treated TiO2/Gr/Hem catalysts showed an inhibition effect for ORR and Ti–C–N materials garnered high catalytic activity compared with Ti–C–N–Fe materials in alkaline media. The higher methanol tolerance and durability of the TiO2/Gr/Hem composite materials during ORR were also confirmed. These results reflected the critical influences of the pyrolysis temperature and the chemically bonded element dopants to be the key factor for ORR.

Electrocatalytic reduction of a coreactant using a hemin–graphene–Au nanoparticle ternary composite for sensitive electrochemiluminescence cytosensing

Xiaoyan Xin,Yiying Yang,Jing Liu,Xiaomeng Wang,Hong Zhou,Bing Yu

A ternary composite of hemin–reduced graphene oxide–Au nanoparticles (H–RGO–Au) with a higher electrocatalytic activity for the reduction of hydrogen peroxide than that of the hemin–graphene component was synthesized and was used as a catalyst to initiate sensitive electrochemiluminescence (ECL) quenching of quantum dots (QDs) due to the electrocatalytic reduction of the coreactant H2O2. Based on this and specifically on the recognition between concanavalin A (Con A) and mannose or N-glycans on the cell surface, an ultrasensitive cytosensor for the detection of 5 cancer cells was first proposed. The designed strategy shows the possibility of expansive application of versatile ECL cytosensing platforms for the detection of many important biomolecules.

Xueying Li,Jing Zhao,Beijiao Cui,Yiming Li,Doudou Zhang,Haiyan Liu,Ligai Bai,Hongyuan Yan,Yong Wang

A poly(hemin-co-C16-co-TEGDA) monolith was prepared using hemin and 1-hexadecene (C16) as monomers; triethylene glycol dimethacrylate (TEGDA) as crosslinking agent; N,N-dimethylformamide and isopropyl alcohol as porogens; and dibenzoyl peroxide and N,N-dimethyl aniline as initiators in 50 × 4.6 mm i.d. stainless steel column. The morphology and structure of the monolith were characterized by scanning electron microscopy and nitrogen adsorption/desorption isotherm measurement, respectively. Compared with poly(C16-co-TEGDA) monolith, poly(hemin-co-C16-co-TEGDA) monolith had better mechanical properties, higher permeability, and better separation ability. The prepared monolith was used to separate not only proteins but also small molecules.
