A facile bioluminescent protocol was developed to detect the total amount of viable Gram-positive bacteria based on a novel antibiotic-affinity strategy. Vancomycin-functionalized magnetic particles were adopted to isolate Gram-positive bacteria from sample matrix, utilizing the five-point hydrogen bonds binding between vancomycin and D-Alanyl- d -Alanine moieties on the cell wall. Then the bioluminescence signal from the bacterial intracellular adenosine triphosphate was collected to quantify the captured bacteria after cells lysis. In this proof-of-principle work, the developed protocol was applied to detect four Gram-positive bacteria, including Staphylococcus aureus , Micrococcus luteus , Bacillus cereus and Streptococcus mutans . It showed a linear range of 1.0 × 10 2 –1.0 × 10 7 CFU mL −1 and a detection limit of 33 CFU mL −1 for the four model bacteria. The whole assay process could be completed within 70 min. Gram-negative bacteria and dead bacteria all showed negligible interference to the detection of the viable Gram-positive bacteria. The method was successfully applied to quantify the amount of viable Gram-positive bacteria in environmental, food and pharmaceutical samples with acceptable recovery values ranging from 72.0% to 120.0%. The protocol possessed numerous advantages such as high sensitivity, low cost, ideal specificity and short detection time. It could also be extended to Gram-negative bacteria detection by utilizing other antibiotics acting on Gram-negative bacteria.

Background Catalpol (CAT), a naturally occurring iridoid glycoside sourced from the root of Rehmannia glutinosa , affects mitochondrial metabolic functions. However, the mechanism of action of CAT against pyrexia and its plausible targets remain to be fully elucidated. Purpose This study aimed to identify the specific targets of CAT for blocking mitochondrial thermogenesis and to unveil the unique biological mechanism of action of the orthogonal binding mode between the hemiacetal group and lysine residue on the target protein in vivo . Methods Lipopolysaccharide (LPS)/ carbonyl cyanide 3-chlorophenylhydrazone (CCCP)-induced fever models were established to evaluate the potential antipyretic effects of CAT. An alkenyl-modified CAT probe was designed to identify and capture potential targets. Binding capacity was tested using in-gel imaging and a cellular thermal shift assay. The underlying antipyretic mechanisms were explored using biochemical and molecular biological methods. Catalpolaglycone (CA) was coupled with protein profile identification and molecular docking analysis to evaluate and identify its binding mode to UCP2. Results After deglycation of CAT in vivo , the hemiacetal group in CA covalently binds to Lys239 of UCP2 in the mitochondria of the liver via an ɛ-amine nucleophilic addition. This irreversible binding affects proton leakage and improves mitochondrial membrane potential and ADP/ATP transformation efficiency, leading to an antipyretic effect. Conclusion Our findings highlight the potential role of CA in modulating UCP2 activity or function within the mitochondria and open new avenues for investigating the therapeutic effects of CA on mitochondrial homeostasis.

ATP citrate lyase (ACLY) is a crucial enzyme in glycolipid metabolism. To date, only its one prodrug bempedoic acid has been approved. The discovery of direct ACLY inhibitors is important for developing new lipid-lowering drugs. Grape seeds, used in functional foods and medicines, have lipid-lowering efficacy. However, it remains unknown whether their components directly inhibit ACLY. To investigate it, a multi-component screening method was used and proanthocyanidin B2 was identified as a novel ACLY inhibitor with an IC 50 value of 2.73 μM. Differential scanning fluorimetry confirmed its directly binding to ACLY. Proanthocyanidin B2 acted as a noncompetitive inhibitor in kinetic studies and bound stably with ACLY in molecular dynamics simulations. In vitro , proanthocyanidin B2 reduced lipid accumulation via inhibiting key transcription factors associated with ACLY. This study revealed a novel ACLY inhibitor contributing to lipid-lowering efficacy of grape seeds and provided a unique scaffold for ACLY drug design.

As a persistent organic pollutant, perfluorooctane sulfonate (PFOS) has a serious detrimental impact on human health. It has been suggested that PFOS is associated with liver inflammation. However, the underlying mechanisms are still unclear. Here, PFOS was found to elevate the oligomerization tendency of voltage-dependent anion channel 1 (VDAC1) in the mice liver and human normal liver cells L-02. Inhibition of VDAC1 oligomerization alleviated PFOS-induced nucleotide-binding domain and leucine-rich repeat protein-3 (NLRP3) inflammasome activation. Cytoplasmic membrane VDAC1 translocated to mitochondria was also observed in response to PFOS. Therefore, the oligomerization of VDAC1 occurred mainly in the mitochondria. VDAC1 was found to interact with the ATP synthase beta subunit (ATP5B) under PFOS treatment. Knockdown of ATP5B or immobilization of ATP5B to the cytoplasmic membrane alleviated the increased VDAC1 oligomerization and NLRP3 inflammasome activation. Therefore, our results suggested that PFOS induced NLRP3 inflammasome activation through VDAC1 oligomerization, a process dependent on ATP5B to transfer VDAC1 from the plasma membrane to the mitochondria. The findings offer novel perspectives on the activation of the NLRP3 inflammasome, the regulatory mode on VDAC1 oligomerization, and the mechanism of PFOS toxicity.

Brassinosteroids are steroidal phytohormones that regulate plant development and physiology, including adaptation to environmental stresses. Brassinosteroids are synthesized in the cell interior but bind receptors at the cell surface, necessitating a yet to be identified export mechanism. Here, we show that a member of the ATP-binding cassette (ABC) transporter superfamily, ABCB19, functions as a brassinosteroid exporter. We present its structure in both the substrate-unbound and the brassinosteroid-bound states. Bioactive brassinosteroids are potent activators of ABCB19 ATP hydrolysis activity, and transport assays showed that ABCB19 transports brassinosteroids. In Arabidopsis thaliana, ABCB19 and its close homolog, ABCB1, positively regulate brassinosteroid responses. Our results uncover an elusive export mechanism for bioactive brassinosteroids that is tightly coordinated with brassinosteroid signaling.

In the present work, strong anodic electrogenerated chemiluminescence (ECL) of Ru(bpy)32+ was observed at a graphene oxide modified glassy carbon electrode (GO/GCE) in the absence of coreactants. The electrocatalytical effect of GO on the oxidation of Ru(bpy)32+ suggested that GO itself can act as the coreactant of Ru(bpy)32+ ECL, which can be used to fabricate the ECL biosensor. Thiol group terminated adenosine triphosphate (ATP) aptamer was immobilized on the GO film via DNA hybridization. When gold nanoparticles/graphene oxide (AuNPs/GO) nanocomposites were modified on the aptamer through the S–Au bond to form a sandwich-like structure, the ECL resonance energy transfer (ECL-RET) could occur between Ru(bpy)32+ and AuNPs/GO nanocomposites, resulting in an apparent decrease of ECL signal. After the ECL sensor was incubated in ATP solution, the AuNPs/GO nanocomposites were released from the electrode due to the specific interaction between aptamer and ATP, leading to the increased ECL signal. On the basis of these results, an ECL aptasensor was fabricated and could be used in the sensitive and selective detection of ATP in the range of 0.02–200 pM with a detection limit of 6.7 fM (S/N = 3). The proposed ECL aptasensor can be applied in the detection of ATP in real samples with satisfactory results.

PRDX3 is a mitochondrial peroxide reductase that regulates cellular redox state. It has been reported that PRDX3 is overexpressed in liver cancer, but how PRDX3 is involved in hepatocellular carcinoma (HCC) tumorigenesis and progression has not been well-characterized. In the present study, we established two stable cell lines by overexpressing or knocking down PRDX3 in HepG2 cells. We found that PRDX3 silencing decreased the growth rate of HepG2 cells and increased mtDNA oxidation. Quantitative proteomics identified 475 differentially expressed proteins between the PRDX3 knockdown and the control cells. These proteins were involved in antioxidant activity, angiogenesis, cell adhesion, cell growth, ATP synthesis, nucleic acid binding, redox, and chaperones. PRDX3 knockdown led to the down-regulation of ATP synthases and the decreased cellular ATP level, contributing to the slow-down of cell growth. Furthermore, silencing PRDX3 enhanced invasive properties of HepG2 cells via TIMP-1 down-regulation and the increased ECM degradation. Taken together, our results indicate that PRDX3 promotes HCC growth and mediates cell migration and invasiveness and is a potential therapeutic target for HCC treatment.

Phytic acid (IP6) is a natural phosphorylated inositol, which is abundantly present in most cereal grains and seeds. This study investigated the effects of IP6 regulation on P-glycoprotein (P-gp) and its potential mechanisms using in situ and in vitro models. The effective permeability of the typical P-gp substrate rhodamine 123 (R123) in colon was significantly increased from (1.69 ± 0.22) × 10–5 cm/s in the control group to (3.39 ± 0.417) × 10–5 cm/s (p < 0.01) in the 3.5 mM IP6 group. Additionally, IP6 can concentration-dependently decrease the R123 efflux ratio in both Caco-2 and MDCK II-MDR1 cell monolayers and increase intracellular R123 accumulation in Caco-2 cells. Furthermore, IP6 noncompetitively inhibited P-gp by impacting R123 efflux kinetics. The noncompetitive inhibition of P-gp by IP6 was likely due to decreases in P-gp ATPase activity and P-gp molecular conformational changes induced by IP6. In summary, IP6 is a promising P-gp inhibitor candidate.

A novel Mg-based defect-modified composite material (MOF-Mg(OH) 2 -HP) was prepared using MOF as a precursor, featuring a unique layered structure with abundant oxygen defects and delocalized electrons on its surface. • MOF-Mg(OH) 2 -HP can be activated by natural light, generating a large amount of reactive species through the “light-Fenton-like” pathway. Superoxide radicals (·O 2 − ) and holes (h + ) are the primary reactive species, which simultaneously oxidize non-reactive organophosphorus compounds and recover orthophosphate (achieving a 100% phosphorus removal rate within 90 min). • It demonstrates exceptional multifunctionality in actual aquaculture wastewater, effectively eliminating 92.84% of Escherichia coli and 98.69% of Chlorella pyrenoidosa , providing a green synergistic solution for integrated water purification and resource recovery strategies. The low-cost and harmless treatment of dairy wastewater is a focal point in agricultural development today. This study innovatively utilizes hydrogen peroxide to modify Mg-MOF-derived oxides, preparing a series of hydrogen peroxide-modified composite materials rich in defects. For the first time, an integrally formed composite material consisting of Mg(OH) 2 with O 2 2− on the outside and MgO on the inside is synthesized. Compared to modified materials prepared using commercially available MgO as the raw material, this material possesses unique Mg(OH) 2 /MgO heterojunctions, a larger specific surface area, abundant oxygen defects, and higher active oxygen content. Starting from the current issues of poor universality, high energy consumption, and difficulties in simultaneous harmless treatment of wastewater in organic phosphorus oxidation materials, we designed an efficient degradation of various organic phosphorus compounds and resource recovery of orthophosphorus using sunlight. This is mainly achieved through the synergistic effect of surface O 2 2− hydrolysis (Fenton-like pathway) and the generation of oxidizing active species through photoexcitation at oxygen vacancies (photocatalytic pathway), resulting in efficient degradation of organic phosphorus and simultaneous recovery (with a removal rate of 100% in 90 min). This material is the first to demonstrate the ability to simultaneously kill 92.84% of Escherichia coli and 98.69% of Chlorella pyrenoidosa during phosphorus removal, proposing a novel collaborative solution for green recovery and harmless treatment of pollutants. A general strategy for the in-situ hydrothermal synthesis of transition metal sulfides functionalized hierarchically porous carbons (FeS 2 -HPC, Co 9 S 8 -HPC, Ni 3 S 2 -HPC, Cu 2 S-HPC, and ZnS-HPC) is proposed using different transition metal chlorides as metal sources and activators, sustainable agroforestry waste biomass ( Citrus sinensis peel sponge layer) as a carbon source, sulfuric acid as a sulfur source, and silica colloid as a hard template. The introduction of both transition metal salts and silica colloids facilitate the adjustment of the pore structure. The added sulfuric acid promotes the degradation of the agroforestry waste biomass, which are conducive to the formation of porous carbons. The in-situ synthesis of transition metal sulfide particles and the sulfur doping improve the conductivity, electrocatalytic and enzyme-like activities of obtained composite materials. The FeS 2 -HPC with multiple enzyme-like activities and the best electrocatalytic activity is applied as a nanozyme electrochemical sensing platform for the simultaneous detection of three ATP metabolites with a limit of detection (LOD) of 0.069 μM for uric acid in a linear range of 0.3–200 μM, a LOD of 0.047 μM for xanthine in 0.3–100 μM, and a LOD of 0.21 μM for hypoxanthine in 0.7–300 μM. This work provides a new idea for the development of transition metal sulfides functionalized porous carbons from agroforestry waste biomass, and a theoretical basis for the fabrication of nanozyme electrochemical sensing platform for health assessment and disease prevention.

Exosomes are extracellular vesicles that inherit molecular markers from their parent cells for the transfer and delivery of membrane and cytosolic molecules between cells. Exosome detection and protein profiling provide noninvasive access to disease diagnosis and treatment. The nanozyme designed in this paper contains polydopamine-functionalization of graphene oxide (rGO@PDA) units that can adsorb ssDNA aptamers and CeO 2 metal oxide nanosheet units with peroxidase activity. As auxiliary regulator, adenosine triphosphate (ATP) enhances peroxidase activity of rGO@PDA@CeO 2 under a wide range of pH (4–7.4), which breaks through the limitation that traditional nanozymes can only exhibit peroxidase activity under acidic conditions. The ssDNA aptamers, which are recognition probes of exosome, can be adsorbed on the surface of rGO@PDA by aromatic stacking, competing with ATP molecules. Thus, the ssDNA aptamer acts as the switch for colorimetric sensor to flexibly regulate the peroxidase activity of the rGO@PDA@CeO 2 under physiological conditions , and realizes sensitive detection of exosomes. This assay achieves a detection linear range of 0.46 × 10 7 –10 8 particle/mL and a limit down to 3.81 × 10 5 particles/mL. In addition, this aptasensor sensitively profiles six exosomal protein across five cell types and clinical samples, suggesting their potential in clinic diagnosis.
