Introduction: Invasive aspergillosis (IA) is a life-threatening disease in immunocompromised individuals, creating an urgent need for rapid, sensitive, and user-friendly methods for early detection of Aspergillus spores in clinical and environmental samples. Methods: In this study, we developed a rapid ultrasonication-based beads beating (USBB) method for cell lysis and fungal DNA release, along with an integrated magnetic beads-based direct amplification (MBDA) method utilizing microfluidic chip technology. Our flexible thin-film microfluidic chip was developed to enable immediate contacting with an ultrasonic oscillator for nucleic acid extraction. Results and discussion: The chip achieved a lysis efficiency of 85.5% for Aspergillus at 10 5 spores per sample. Magnetic beads releasing Fe ions at 75-860 ng/mL allowed high-efficiency direct PCR amplification. Complete transfer of the nucleic acid extract into PCR amplification led to a 100-fold improvement in detection sensitivity. The integrated USBB-MBDA system completed the entire workflow from sample-in to result-out within 35 min (5 min fungal DNA release and 30 min TaqMan assay). Using this approach, we achieved simultaneous triplex detection of Aspergillus fumigatus, Aspergillus flavus , and Aspergillus niger , with a sensitivity as low as 10 spores per test. Validation on 37 clinical samples showed complete concordance with MALDI-TOF mass spectrometry (MS). This study establishes an integrated nucleic acid extraction and magnetic bead–enabled direct amplification strategy, providing a versatile approach for the development of automated analytical platforms with broad applicability to in vitro diagnostics.

The in situ fabricated perovskite nanocrystals (PNCs) obtained through spin-coating a precursor solution are suitable candidates to achieve efficient perovskite light emitting diodes (PeLEDs). In this work, the solvent effects of on-chip crystallization are investigated by correlating the nucleation and growth process of in situ fabricated formamidinium lead iodide (FAPbI 3 ) nanocrystals with their optical and electronic properties. The FAPbI 3 nanocrystals obtained from a precursor solution in γ-butyrolactone (GBL) are smaller than those obtained from N , N -dimethylformamide and dimethyl sulfoxide, and the relatively weak coordination between GBL and the precursor molecules enables reduced defect states in the resulted PNCs with enhanced photoluminescence properties. A modified LaMer model is proposed to describe the solvent effects in the on-chip crystallization process. Based on these understandings, red emissive FA 0.87 Cs 0.13 PbI 3 nanocrystal films with absolute photoluminescence quantum yields up to 70% are realized. Finally, an efficient PeLED with maximum luminance of 218 cd m −2 and peak external quantum efficiency of 15.8% is achieved with good reproducibility.

Background Sepsis, a systemic inflammation syndrome initiated by infection, poses significant challenges due to its intricate pathophysiology. T cells play a crucial role in combating infections during sepsis. Despite previous observations indicating T cell dysfunction in sepsis, reliable in-vitro detection methods were lacking, and the factors influencing these impairments remained unclear. Methods We developed a novel method using the D 4 -Chip to assess sepsis T cell migration function. This microfluidic platform enabled precise analysis of migration function under controlled conditions. Additionally, We explored the impact of the plasma microenvironment on T cell behavior, along with the redox environment in sepsis, and assessed the potential efficacy of Mitoquinone mesylate (MitoQ), a mitochondrial-targeted drug. Results Our findings revealed impaired migration function in sepsis T cells compared to healthy controls. Interestingly, sepsis plasma enhanced the migration of healthy T cells, yet incubation with healthy plasma did not fully restore migration impairments in sepsis T cells. Subsequent investigations uncovered a significant increase in NADH/NAD+ levels in sepsis T cells, with healthy T cells exposed to various sepsis plasma conditions also showing elevated NADH/NAD+ levels. Importantly, MitoQ normalized abnormal intracellular NADH/NAD+ levels and enhanced the migration ability of T cells. Conclusions Short-term incubation with sepsis plasma does not directly inhibit T cell migration but instead affects T cell function by disrupting the intracellular redox environment. Improving the intracellular redox environment of sepsis patients contributes to restoring impaired migration and proliferation, with MitoQ demonstrating therapeutic potential.

First synthesis of Mn-CeO 2 @ZnGa₂O₄:Cr³⁺, Eu³⁺ composite. • Trimodal sarcosine detection using phosphorescence, peroxidase mimicry, and photothermal TMB. • Smartphone-based mobile tool enables on-site sarcosine detection. • Paper-based chip sensor simplifies sarcosine detection, making it facile and convenient. Prostate cancer (PCa) is the second most common cancer in men, while sarcosine levels have profound implications for the state of prostate cancer. Herein, we have designed a smart paper-chip sensor based on Mn-CeO₂ nanozyme@persistent luminescent nanoparticles (PLNPs) for trimodality fluorescence, colorimetric, and photothermal detection of sarcosine. The PLNPs were prepared as ZnGa₂O₄: 0.5% Cr 3+ , 1.5% Eu 3+ and then integrating Mn-CeO₂ nanozyme. The composite was integrated onto a polypropylene (PP) substrate as the paper chip. In pa1rticular, PLNPs exhibited the afterglow emission for eliminating the interference from the autofluorescence of biological samples; In the presence of both sarcosine and sarcosine oxidase, red phosphorescence is restored, for phosphorescence detection. The nanozyme of Mn-doped CeO₂ exhibit enhanced peroxidase activity, rapidly converting colorless 3,3′,5,5′-tetramethylbenzidine (TMB) into a blue form as oxidized TMB, which also possesses photothermal conversion capabilities, enabling both colorimetric and photothermal detection. Using a smartphone, this sensor enables on-site detection of sarcosine in serum with a detection range of 0–10 μM with extremely low limits of detection (LOD) as 1.88 μM for fluorescence; 0.08 μM for colorimetry; and 1.88 μM for photothermal mode, with all RSD below 5%. Overall, we have successfully designed for the detection of sarcosine in serum. The sensor's low cost, high sensitivity, and portability have been validated, thereby improving treatment success rates and survival rates for patients with early-stage prostate cancer. Scheme 1. Schematic illustration of a smartphone-based Mn-CeO₂@persistent luminescence nanoparticles (PLNPs) paper-based sensor for triple-mode detection of sarcosine. ZnGa₂O₄: Cr 3+ , Eu 3+ (ZCEGO) was fabricated as PLNPs featuring red emission at 700 nm. The PLNPs were surface-functionalized with Mn-CeO₂ nanozymes as peroxidase mimics, and the resultant nanocomposite was further incorporated onto a paper-based substrate. In the presence of sarcosine and sarcosine oxidase (SOX), H₂O₂ is generated. In Zone 1 (yellow section), the PLNPs are exposed, enabling phosphorescence detection. Meanwhile, in the presence of TMB, TMB in Zone 2 (white section) is oxidized to oxTMB, enabling colorimetric detection. The oxTMB can be used for photothermal detection upon irradiation with an 808 nm laser. Quantitative analysis of sarcosine is achieved using a smartphone. Based on the confined flow - dual-network fixation mechanism, a self-adaptive composite phase change material was prepared. • In the chip thermal management, this composite phase change material could reduce the CPU temperature by up to 31.25 °C. • The composite phase change material has self-adaptive capability, reducing the interface thermal resistance by up to 78.56%. With the continuous trend toward high integration, chips are increasingly subjected to high heat-flux densities. However, excessive heat-flux density can induce chip overheating and warpage, increase local interfacial thermal resistance, and ultimately raise the risks of heat-dissipation failure and performance degradation. Therefore, thermal management has become a critical bottleneck restricting the further development of advanced chips, whereas conventional thermal interface materials (TIMs) still struggle to simultaneously accommodate chip warpage and maintain structural stability. To address this challenge, this study proposes a self-adaptive thermal-interface strategy based on a confined-flow and dual-network fixation mechanism. By constructing a calcium chloride tetrahydrate (CaCl 2 ·4H 2 O)/expanded graphite (EG)/in situ-grown silicon carbide nanowire (SiCnws) system, a high-efficiency heat-dissipating composite phase change material (CPCM) was developed, enabling simultaneous warpage conformability and structural stability. When applied to central processing unit (CPU) thermal management, the CPCM achieved a maximum core-temperature reduction of 31.25 °C. After 300 thermal cycles, the maximum fluctuation in the CPU core temperature was only 10.9 °C. Characterization results showed that, upon heating, the CPCM softened and conformed tightly to curved copper sheets, thereby reducing local thermal resistance. Interfacial thermal resistance measurements indicated that the adapted interface exhibited a resistance of 0.215 cm 2 ·K·W −1 , corresponding to a 78.56% reduction relative to the non-adapted state. This study provides a promising technical pathway and theoretical guidance for addressing chip warpage and high-heat-flux dissipation challenges.

Microfluidic
chips are promising for on-site analysis in medical
diagnostics and food safety because they enable miniaturized, integrated
sample processing. However, the analysis of complex, salt-rich matrices
of biological and food samples remain challenging for conventional
closed chips and electrokinetic pretreatment methods. Here, we developed
an open, reconfigurable electrophoresis chip based on liquid plasticine,
in which a moving neutralization reaction electrophoresis system (MNRES)
was implemented for high-salt sample pretreatment. Liquid plasticine
enabled facile construction of pump-free, valve-free, optically transparent,
and cuttable open reservoirs and channels. In the MNRES, OH – and H + generated by water electrolysis reacted with NH 4 + and CH 3 COO – , producing
moving neutralization reaction boundaries accompanied by dynamic electric
field and pH gradients. These two coupled gradients enabled salt removal,
field amplified stacking, and meanwhile the pH gradient induced protein
focusing, thereby allowing simultaneous enrichment and purification
of proteins and anionic species. It is demonstrated that proteins
and anionic species in 0.9% NaCl solution can be effectively separated
and enriched by approximately 10-fold. Moreover, the MNRES-pretreated
analyte zones can be directly monitored by smartphone-based online
colorimetry or transferred to liquid-plasticine microreactors for
offline chromogenic assays. This work provides a flexible open-fluidic
strategy for integrated electrokinetic pretreatment and visual detection
of complex high-salt samples.

Foodborne pathogens endanger public health and rapid, sensitive, and accurate detection of them is vital. Portable, highly integrated detection devices have great application prospects in the screening and analysis of foodborne pathogens. In this study, a fully automated detection device based on a fully enclosed microfluidic chip cartridge was successfully designed. This device integrates multiple functions, including nucleic acid extraction, reagent preparation, LAMP reaction, and signal detection. By simply adding a sample, it can simultaneously detect four types of foodborne pathogens, making it advantageous for the analysis of complex samples and improving detection accuracy. Additionally, freeze-dried reagents are integrated into the fully enclosed microfluidic chip cartridge, which allows the reagents to be transported and stored at room temperature, greatly reducing the cost of detection. It has been successfully applied in actual samples contaminated with multiple foodborne pathogens and has excellent stability. The entire detection process can be completed in 45 minutes, with a sensitivity of approximately 500 CFU mL−1. Therefore, the automated microfluidic device would be adequate for point-of-care testing (POCT) with high simplicity and high speed, providing an advanced genetic analysis microsystem for foodborne pathogen detection.

Carbon dioxide (CO 2 ) emissions pose significant threats to human health and the environment. Developing highly sensitive CO 2 sensors operating at room temperature is critical for environmental and industrial applications. This study introduces a room-temperature CO 2 sensing platform based on polyethyleneimine/polyethylene glycol-functionalized zinc oxide nanowires (PEI/PEG-ZnONWs) composites. Under ambient conditions (25 °C, 45% relative humidity), the sensor demonstrates a high response of 67% to 5000 ppm CO 2 , with rapid response and recovery times of 13 s and 61 s, respectively. Notably, ultraviolet (UV) (365 nm) activation enhances photogenerated carrier separation, significantly accelerating gas-sensitive kinetics and reducing response/recovery times to 5 s and 19 s, respectively. The sensor demonstrates excellent selectivity, stability, and reproducibility. Mechanistic studies attribute this enhancement to the amine-rich active sites introduced by PEI/PEG functionalization and the high surface area of ZnONWs, which facilitate gas adsorption/desorption dynamics and optimize charge transport through conductive pathways. Finally, the platform enables real-time monitoring of dynamic CO 2 changes in plants, distinguishing respiration from photosynthesis. These findings highlight the potential of PEI/PEG-ZnONWs sensors for CO 2 detection in industrial safety, environmental surveillance, and healthcare.

Simultaneous analysis of multiple protein and nucleic acid biomarkers in one clinical sample can provide clinically valuable results for the diagnosis, monitoring, and management of diseases. However, current diagnostic platforms primarily rely on the independent detection of proteins and nucleic acids, which is time-consuming and labor-intensive. In this study, we propose a microfluidic chip system that integrates microfluidic molecular Point-of-Care Testing (POCT) technology and microfluidic homogeneous chemiluminescence analysis (CLIA) technology for simultaneous detection of protein and nucleic acid biomarkers of ovarian cancer (OC). The integrated system shows good detection limits of 10 fM for miR-92a and 15 pM for HE4. In primary clinical tests with human samples, the diagnostic sensitivity and specificity of the simultaneous detection of two biomarkers (miR-92a and HE4) for OC were 100 % and 96.1 %, respectively, and the area under the curve (AUC) was 0.996. Therefore, the combined evaluation of multiple biomarkers of different dimensions can improve the specificity and sensitivity of diagnosis. This system can also be applied to other tumor markers and biomarkers in other fields of diseases, offering a broad spectrum of diagnostic capabilities.

The rapid advancement of artificial intelligence (AI) technology is creating new requirements for supply voltage stability and power conversion efficiency, necessitating electrolytic capacitors with conductive polymer cathode films. A study examines how different doping anions, pH levels, and current densities affect the constant-current electropolymerization of poly(3,4-ethylenedioxythiophene) (PEDOT) on poly(3,4-ethylenedioxythiophene):poly(styrene sulfonate) (PEDOT:PSS) substrates. Highly doped PEDOT films with smooth surfaces and a conductivity of 313 S cm −1 are obtained at pH 2 and 0.5 mA cm −2 using dodecyl benzene sulfonate (DBS − ) as a doping anion. A PEDOT:PSS conductive substrate is created on the dielectric surface of anodized aluminum foil. Low current density and then high current density electropolymerization of PEDOT cathodes on substrate allow for more efficient capacitors with high capacitance and low leakage current. Based on this, the chip capacitor is obtained with a capacitance of 80 μF cm −2 level, a loss of 0.65 % at 100 Hz, an equivalent series resistance (ESR) of 4.16 mΩ at 100 kHz, and a leakage current of 0.8 μA at 16 V. This work is significant for the research of ultra-low loss and ESR chip aluminum solid electrolytic capacitors.

Extracellular vesicles (EVs) carry disease-specific molecular profiles, demonstrating massive potential in biomarker discovery. In this study, we developed an integrated biochip platform, termed EVID-biochip (EVs identification and detection biochip), which integrates in situ electrochemical protein detection with on-chip antifouling-immunomagnetic beads modified with CD81 antibodies and zwitterion molecules, enabling efficient isolation and detection of neuronal EVs. The capability of the EVID-biochip to isolate common EVs and detect neuronal EVs associated with Parkinson's disease in human serum is successfully demonstrated, using the transmembrane protein L1-cell adhesion molecule (L1CAM) as a target biomarker. The EVID-biochip exhibited high efficiency and specificity for the detection of L1CAM with a sensitivity of 1 pg/mL. Based on the validation of 76 human serum samples, for the first time, this study discovered that the level of L1CAM/neuronal EV particles in serum could serve as a reliable indicator to distinguish Parkinson's disease from control groups with AUC = 0.973. EVID-biochip represents a reliable and rapid liquid biopsy platform for the analysis of complex biofluids offering EVs isolation and detection in a single chip, requiring a small sample volume (300 µL) and an assay time of 1.5 h. This approach has the potential to advance the diagnosis and biomarker discovery of various neurological disorders and other diseases.

As thiocyanate (SCN − ) acts as an important biomarker in human health assessment, there remains an urgent need to realize rapid and reproducible analysis of SCN − in body fluids. Here, a droplet microfluidic device has been designed and fabricated for SCN − detection in real human serum and saliva using the surface enhanced Raman scattering (SERS) technique. Only a few minutes are needed for the whole detection process which simply cost a few microliters of real sample. Gold@silver core–shell nanorods (Au@Ag NRs) with a large SERS enhancement factor were selected to capture SCN − ions in body fluids. The intensity of SERS peak at around 2100 cm −1 , which originates from the –C≡N stretching mode, was used to indicate the concentrations of SCN − ions. Importantly, by generating a droplet environment for mixing reagents and acquiring signals, this microfluidic platform possesses the advantages of an improved reproducibility and reduced time consumption. For practical applications, the SERS-microfluidic system is capable to achieve rapid analysis of SCN − in the presence of human serum, which is very important for realizing the detection in real biological samples. Additionally, SCN − in saliva samples was detected in the SERS-microfluidic chip and the results provide useful information for distinguishing between smokers and nonsmokers.
