Gallium-68-Labelled Indocyanine Green as a Potential Liver Reserve Imaging Agent

Yuxiao Xia,Li Zhang,Yanhong Zhao,Xiangdong Liu,Liang Cai,Lin Liu,Yue Chen,Wei Zhang

Objective. This work evaluated the potential of 68Ga-labelledNOTA-ICG (1,4,7-triazacyclononane-1,4,7-triacetic acid indocyanine green) for liver reserve imaging. Methods. To determine the optimal conditions for generating 68Ga-NOTA-ICG, various reaction parameters were implemented. Quality control analysis was performed using different chromatography techniques. The in vitro and in vivo stability was also measured at specific time points. The radioactivity ratio between n-octanol and water was determined to evaluate the water solubility of 68Ga-NOTA-ICG. The plasma-protein binding rate of the labelled compound was determined by the methanol method. The biodistribution and imaging findings were evaluated in normal animals at different time points after injection. A preliminary imaging evaluation was performed using an animal model of hepatic ischaemia-reperfusion injury, which was confirmed by pathology. Results. 68Ga-NOTA-ICG was prepared with very high radiochemical purity (>98%) by reacting at 90°C for 10 min at pH = 3.5∼4.0, with excellent stability in vivo and in vitro (>95% 3 h postpreparation). The in vitro plasma-protein binding rate of 68Ga-NOTA-ICG was 13.01 ± 0.7%, and it showed strong water solubility . We found that in addition to excretion through the biliary tract and intestines, 68Ga-NOTA-ICG can be excreted through the urinary tract. The image quality of 68Ga-NOTA-ICG was very high; imaging agent retained in the area of liver injury could clearly be observed. Conclusion. This is the first report on a 68Ga-labelled NOTA-ICG fragment for liver reserve function studies. This complex has promise as a candidate agent for liver reserve imaging.

NIR Light-Triggered Chemo-Phototherapy by ICG Functionalized MWNTs for Synergistic Tumor-Targeted Delivery

Lu Tang,Aining Zhang,Yijun Mei,Qiaqia Xiao,Xiangting Xu,Wei Wang

The combinational application of photothermal therapy (PTT), chemotherapy, and nanotechnology is a booming therapeutic strategy for cancer treatment. Multi-walled carbon nanotube (MWNT) is often utilized as drug carrier in biomedical fields with excellent photothermal properties, and indocyanine green (ICG) is a near-infrared (NIR) dye approved by FDA. In addition, ICG is also a photothermal agent that can strongly absorb light energy for tumor ablation. Herein, we explored a synergistic strategy by connecting MWNT and a kind of ICG derivate ICG-NH2through hyaluronic acid (HA) that possesses CD44 receptor targeting ability, which largely enhanced the PTT effect of both MWNT and ICG-NH2. To realize the synergistic therapeutic effect of chemotherapy and phototherapy, doxorubicin (DOX) was attached on the wall of MWNT via π–π interaction to obtain the final MWNT-HA-ICG/DOX nanocomplexes. Both in vitro and in vivo experiments verified the great therapeutic efficacy of MWNT-HA-ICG/DOX nanocomplexes, which was characterized by improved photothermal performance, strengthened cytotoxicity, and elevated tumor growth inhibition based on MCF-7 tumor models. Therefore, this synergistic strategy we report here might offer a new idea with promising application prospect for cancer treatment.Keywords:multi-walled carbon nanotube;photothermal therapy;indocyanine green;synergistic strategy;cancer treatment;targeted drug delivery

Oligopeptide modified ICG/Cisplatin@CaCO3 for targeted osteosarcoma NIR-II imaging guided multimodal therapy

Li-chen Ji,Jia-qing Huang,Xu-gang Zhong,Qi-hong Sun,Yong Fan,Li Yin,Qiong Zhang,Yu Cai,Zeju He

Osteosarcoma, as the most prevalent primary malignant bone tumor, has consistently shown suboptimal clinical treatment outcomes. In this manuscript, we proposed an efficacious and novel phototherapy strategy for osteosarcoma based on a photoresponsive near-infrared (NIR) material, CaCO 3 -ICG-DDP-PEG-PT (CIDP-PT), which contained calcium carbonate (CaCO 3 ) and Cisplatin (DDP) via the gas dispersion method, and indocyanine green (ICG) was encapsulated through a nanoprecipitation reaction by employing DSPE-PEG 2000 -COOH to enhance the stability and hydrophilicity. The targeting oligopeptide (PT) for osteosarcoma cells was inserted into the outer membrane of nanoparticles, thereby further enhancing the tumor-targeting capability through receptor-mediated binding. The prepared CIDP-PT nanoparticles exhibited high targeting efficiency and multifunctionality. In vitro and in vivo experiments demonstrated that CIDP-PT significantly enhanced the therapeutic efficacy for osteosarcoma via the NIR-II photoresponsive and tumor-targeting approach. It achieves efficient tumor cell eradication through multiple modalities while maintaining an exceptional level of biocompatibility. Hence, the CIDP-PT nanocarriers possess considerable practical value for future clinical applications in adjunct osteosarcoma phototherapy.

Rare-Earth Cerium-Coordinated ICG Nanoprobe for Tumor Hypoxia Relief and Intensified Photodynamic Therapy

Xiaohang Liu,Zhiyang Fan,Liya Xie,Wei Zang,Li Li,Yueyang He,Dongfang Ding,Jianwu Chen,Yun Zhang,Xiaolong Liu,Yao Huang,Yang Li

Photodynamic therapy (PDT) utilizing organic photosensitizers like indocyanine green (ICG) faces several intrinsic limitations. These challenges include a propensity to aggregate, insufficient stability, and low intersystem crossing (ISC) efficiency that yields inadequate reactive oxygen species (ROS). Furthermore, the tumor microenvironment imposes additional restrictions on its efficacy. To address these challenges, we implemented a cetyltrimethylammonium bromide (CTAB)–templated metal coordination approach and rationally designed cerium(IV)-coordinated, self-assembled ICG nanoparticles (CINPs). Cooperative coordination between Ce 4+ and ICG, combined with hydrophobic interactions, significantly enhances ICG stability. It also optimizes the energy gap between the lowest singlet excited state (S 1 ) and the lowest triplet state (T 1 ), thereby promoting ISC and equipping the system with robust ROS-generating capacity. Moreover, Ce 4+ exhibits dual catalytic activity, on the one hand catalyzing the decomposition of endogenous hydrogen peroxide (H 2 O 2 ) to generate oxygen and alleviate tumor hypoxia, and on the other hand oxidizing and depleting intracellular glutathione (GSH) to weaken antioxidant defenses. In a hepatocellular carcinoma model, CINPs harnessed efficient ROS production to induce mitochondrial dysfunction, lipid peroxidation (LPO), and DNA strand breaks, which collectively activated multiple cell death pathways and significantly suppressed tumor growth. Unlike nanoplatforms that require elaborate designs, complex compositions, and fine chemical synthesis, the nanoparticles developed here are assembled from a small set of reliable, clinically established materials and can be rapidly formed through a simple process in less than 30 minutes. Despite the streamlined preparation, CINPs effectively remodel the tumor microenvironment, induce vigorous ROS generation during treatment, and achieve potent antitumor activity via oxidative stress-related mechanisms across multiple pathways, highlighting strong translational potential and broad prospects for clinical application.

Nanoformulation-enabled CDDP/ICG combination for synergistic chemotherapy and photothermal therapy of ovarian cancer

Qidan Huang,Jingsong Huang,Ting Deng,Ping Yang,Jieping Chen,Wen Zhang,He Huang,Weimin Liu

Ovarian cancer (OC) remains a highly lethal gynecologic malignancy, with platinum (Pt)-based chemotherapy facing challenges from drug resistance and systemic toxicity. In this study, we developed a silica-based nanoparticle system, termed SiO₂@PEG-ICG&CDDP, to co-deliver cisplatin (CDDP) and indocyanine green (ICG) for synergistic chemotherapy and photothermal therapy (PTT) against OC. The nanoplatform encapsulates cisplatin (CDDP) and indocyanine green (ICG), leveraging the enhanced permeability and retention (EPR) effect for tumor accumulation. Under 808 nm near-infrared (NIR) irradiation, ICG-mediated photothermal heating not only induces tumor ablation but also enhances cellular uptake of CDDP and suppresses DNA repair mechanisms. Concurrently, CDDP promotes apoptosis via the formation of platinum–DNA adducts, disrupting DNA replication and transcription. In vitro and in vivo evaluations demonstrated that this combinatory approach effectively reverses CDDP resistance and significantly suppresses tumor growth, while minimizing systemic side effects. Collectively, SiO₂@PEG-ICG&CDDP represents a promising nanotherapeutic strategy to augment the efficacy of platinum-based chemotherapy in ovarian cancer through PTT–chemotherapy synergy.

Preparation of Photoresponsive Doxorubicin/ICG Microfluidic Nanoparticles and Their Enhanced Anti-B16F10 Melanoma Activity

Xia Cao,Zhangxintian Ding,Yize Lv,Xuedi Weng,Hongran Huang,Jinghan Li,Ximing Xu,Qilong Wang

The clinical treatment of melanoma is severely limited by the rapid development of intrinsic and acquired resistance to conventional chemotherapeutic drugs, such as doxorubicin (DOX), as well as their severe, dose-limiting systemic toxicities. To address this issue, we developed a novel nanoplatform for near-infrared (NIR)- activatable nanoplatform leveraging microfluidics as a precision fabrication tool to overcome the limitations of conventional nanocarrier synthesis, including poor batch-to-batch reproducibility, heterogeneous size distribution, and suboptimal drug co-loading. A photocrosslinkable copolymer, Pluronic F127 diacrylate (F127DA), was synthesized and processed using a custom-designed dual-zone microfluidic chip that integrates sequential flow-focusing and turbulent mixing. This approach enabled the one-step, highly controlled co-encapsulation of DOX and indocyanine green (ICG) into monodisperse F127DA nanoparticles (43.9 ± 0.6 nm, polydispersity index: 0.18) with superior encapsulation efficiency (DOX: 60.4%) and drug loading (12.8%). Upon NIR irradiation, the resulting DOX/ICG@F127DA nanoparticles mediated synergistic chemo-photothermal-photodynamic therapy, as evidenced by potent cytotoxicity (melanoma cell viability reduced to 35%), a significant 3.1-fold increase in reactive oxygen species (ROS) generation, and effective inhibition of cell migration (wound closure rate of 17.2% compared to 21.7% with free DOX). Mechanistically, the NIR-triggered ROS burst induced mitochondrial-mediated apoptosis by dysregulating the Bax/Bcl-2 ratio (increased by 2.7-fold), culminating in an apoptosis rate of up to 68.3%. Collectively, this work establishes microfluidics as a robust and scalable platform for engineering complex nanomedicines and delineates the “NIR light–ROS–Bax/Bcl-2” axis as a critical molecular pathway for overcoming chemoresistance in melanoma. Ning Ding,Bo Zhang,Imran Mahmood Khan,Mingwei Qin,Shuo Qi,Xiaoze Dong,Zhouping Wang,Junsong Yang

The prevalence of pathogenic bacterial infections with high morbidity and mortality poses a widespread challenge to the healthcare system. Therefore, it is imperative to develop nanoformulations capable of adaptively releasing antimicrobial factors and demonstrating multimodal synergistic antimicrobial activity. Herein, an NIR-activated multifunctional synergistic antimicrobial nanospray MXene/ZIF-90@ICG was prepared by incorporating ZIF-90@ICG nanoparticles onto MXene-NH2 nanosheets. MXene/ZIF-90@ICG can on-demand release the antimicrobial factors MXenes, ICG, and Zn2+ in response to variations in pH and ATP levels within the bacterial infection microenvironment. Under NIR radiation, the combination of MXenes, Zn2+, and ICG generated a significant amount of ROS and elevated heat, thereby enhancing the antimicrobial efficacy of PDT and PTT. Meanwhile, NIR excitation could accelerate the further release of ICG and Zn2+, realizing the multimodal synergistic antibacterial effect of PDT/PTT/Zn2+. Notably, introducing MXenes improved the dispersion of the synthesized antimicrobial nanoparticles in aqueous solution, rendering MXene/ZIF-90@ICG a candidate for application as a nanospray. Importantly, MXene/ZIF-90@ICG demonstrated antimicrobial activity and accelerated wound healing in the constructed in vivo subcutaneous Staphylococcus aureus infection model with NIR activation, maintaining a favorable biosafety level. Therefore, MXene/ZIF-90@ICG holds promise as an innovative nanospray for adaptive multimodal synergistic and efficient antibacterial applications with NIR activation.

The synthesis of core–shell Cu9S5@mSiO2–ICG@PEG–LA for photothermal and photodynamic therapy

Na An,Ying Wang,Meng Li,Huiming Lin,Fengyu Qu

Recently, with strong NIR absorption, CuSx nanomaterials have been considered as potential photothermal and photodynamic anticancer agents. Herein, Cu9S5 nanoparticles were synthesized as the core and coated with mesoporous silica to construct the core–shell nanostructure (Cu9S5@mSiO2). The as-synthesized Cu9S5@mSiO2 nanomaterial possesses uniform sphere morphology, about 50 nm in size. Considering its ample porous structure, Cu9S5@mSiO2–NH2 was used as a nanocarrier to demonstrate the high loading efficiency of the photosensitizer indocyanine green (ICG). Under 808 nm NIR light illumination, the temperature rises to 55.8 °C (3 W cm−2), and the high photo-thermal converter is ascribed the combined action of Cu9S5 and ICG. Moreover, Cu9S5@mSiO2–ICG also reveals the illumination-dependent ROS generation. In addition, polyethylene glycol (PEG) and lactose acid (PEG–LA) was prepared and grafted outside Cu9S5@mSiO2–ICG to further guarantee the targeting and cell uptake performance. Under the synergistic effect of PTT and PDT, the Cu9S5@mSiO2–ICG@PEG–LA nanocomposite shows enhanced cytotoxicity to human hepatoma cells (HepG2), making it a promising candidate in nanomedicine as an antitumor agent.

Toxicity-shielded Ag-Pt-ICG nanocomposite with photothermal-enhanced ionic release for lung cancer therapy

Min Gao,Gege Li,Xiaoran Zheng,Yang Yang,Hai Zhong,Yunlei Zhang,Xiuwei Zhang

Background Silver nanoparticles (AgNPs) show antitumor potential but are limited by off-target oxidative toxicity. By integrating platinum nanoparticles (PtNPs) with indocyanine green (ICG)-mediated near-infrared (NIR) responsiveness, we developed an Ag-Pt-ICG (API) nanocomposite for fluorescence imaging and combined lung cancer therapy with enhanced therapeutic efficacy. Methods Ultrasmall Ag-Pt nanocomposites were synthesized via a seeded-growth strategy and subsequently functionalized with peptides to yield the intermediate conjugate AP-peptide (AP/PP). The photosensitizer indocyanine green bearing an NHS-ester group (ICG-NHS) was conjugated to the nanocomposites via peptide linkage, yielding API constructs. Comprehensive physicochemical characterization, including morphology, particle size, elemental composition, and colloidal stability, was conducted. We evaluated biosafety, and antitumor performance through a series of in vitro and in vivo assays. Results The API-based nanocomposites possessed typical nanoscale morphology and showed superior dispersion stability in serum-supplemented DMEM compared with PBS. Upon 808 nm NIR light irradiation, these nanocomposites achieved efficient photothermal conversion, excellent photothermal cycling stability, and NIR-triggered ROS production, endowing them with combined photothermal/photodynamic therapeutic effects. Under acidic conditions at pH 6.5, the nanocomposites could release silver ions into the surrounding medium, and 808 nm laser irradiation further accelerated silver ion release relative to the control group. In vitro results revealed that combined treatment with API nanocomposites and 808 nm NIR irradiation significantly suppressed the viability of A549 cells and elevated intracellular ROS levels. In vivo experiments confirmed that the combined treatment of API nanocomposites with 808 nm NIR laser exerted prominent tumor growth inhibition and alleviated tumor burden in tumor-xenografted mice compared with control groups. Forty-eight-hour in vivo fluorescence imaging confirmed the effective tumor accumulation of the nanocomposites. The fluorescence signal gradually decreased at later time points, suggesting time-dependent elimination while maintaining detectable tumor retention during the observation period. Furthermore, preliminary biosafety assessment confirmed no evident acute systemic toxicity under the tested conditions, which was evidenced by stable body weight changes, normal liver and renal function-related serum biochemical parameters, as well as hematoxylin-eosin staining results of major vital organs. Collectively, all these findings demonstrate that the as-prepared API nanocomposites are promising multifunctional nanoplatforms for NIR-activated combined tumor therapy with satisfactory biocompatibility. Conclusion The NIR-activated API hybrid nanoplatform synergizes photothermal-photodynamic therapy with Ag-induced cytotoxicity while suppressing off-target ROS toxicity via Pt-mediated antioxidant activity. This approach offers a clinically translatable strategy to enhance both the therapeutic efficacy and safety profile of metal-based nanotherapeutics.
