Bone homeostasis regulation by in vivo generation of osteoclast-derived apoptotic bodies using targeted self-assembly

Zhang Peng,Chen Dinghao,Fang Yu,Li Sangshuang,Zhou Ziao,Lu Huanfen,Cheng Jianjun,Wang Huaimin

Osteoporosis therapies remain limited by non-specific osteoclast inhibition, which disrupts physiological bone remodeling and increases fracture risk. Here we show that selectively targeting mature osteoclasts via enzyme-responsive nanoparticles could eliminate pathological bone resorption while generating apoptotic bodies (ABs) to promote regeneration. We design OsteoSAVE, a peptide-based nanoparticle integrating a cathepsin K (CTSK)-cleavable linker, a bone-targeting motif, and a self-assembling biphenyl core. Upon CTSK-mediated hydrolysis, OsteoSAVE transforms into nanofibers within osteoclast lysosomes, inducing apoptosis and releasing ABs enriched with PDGF-BB. Mechanistic studies confirm CTSK-selective cleavage and charge-driven cellular specificity. In ovariectomized mice, OsteoSAVE restores bone density by coupling osteoclast elimination with ABs-mediated osteogenesis via PI3K/AKT activation in mesenchymal stem cells. This work establishes enzyme-triggered supramolecular self-assembly as a paradigm for dual-action therapies that recalibrate bone homeostasis without disrupting physiological processes.

A novel dual-mode Fluorescence/Colorimetric Aptasensor Based on NH2-MIL-88B(Fe)/CDs-Apt for Kanamycin Detection

Hou Huijing,Luo Mingming,Li Fang,Tan Yijun,Liu Ruowei,Xu Longping,Feng Yanwen,Guo Xiangli,Wu Zijian

Kanamycin (KANA) is a veterinary drug residue that is highly scrutinized in food safety testing. In this study, a metal-organic framework-based aptamer-nanoenzyme (NH 2 -MIL-88B(Fe)/CDs-Apt) was introduced for the dual-mode detection of KANA. NH 2 -MIL-88B(Fe) catalyzes the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) by H₂O₂ to form blue ox-TMB, thereby enabling colorimetric detection. As a fluorescence quencher, NH 2 -MIL-88B(Fe) quenches the fluorescence of CDs via photogenerated electron transfer (PET) or fluorescence resonance energy transfer (FRET) mechanisms. The detection limits were 0.716 fM (fluorescent) and 0.709 fM (colorimetric), respectively. The proposed aptasensor was successfully applied to the detection of KANA in eggs, honey, and lake water. This aptasensor demonstrates high sensitivity and accuracy, as well as broad application potential.

Synergistic action of naringenin and amikacin in the treatment of avian colibacillosis

Yi Lankun,Chen Xu,Cao Mingze,Liu Qing,Huang Yang,Pang Jianing,Wang Zeyu,Guan Ding,Ma Tenghe,Zhu Zhen,Wan Yongbiao

Background Avian colibacillosis is an infectious disease caused by pathogenic Escherichia coli (E. coli) and is one of the significant causes of economic losses in the global poultry industry. The overuse of antibiotics has led to the emergence of resistant strains, and the presence of antibiotic residues in chickens and eggs poses a threat to human health. Therefore, exploring safe adjuvants to enhance the efficacy of existing antibiotics is one of the critical issues in poultry farming and public health research. Naringenin (NG), a naturally occurring flavonoid, has been extensively studied as a therapeutic agent for various diseases, but its efficacy in combination with antibiotics for treating avian colibacillosis has not yet been explored. Results In this study, we established a model of avian colibacillosis and treated it with NG and amikacin (AMK) alone or in combination. Compared with monotherapy, the combination of NG and AMK significantly decreased bacterial loads in the heart, liver, spleen, lung, and kidney by 5.0–6.0 log 10 CFU/g ( p  < 0.01). Histopathological analysis revealed that the combination treatment markedly alleviated pathological damage in the lung and duodenum compared to the AMK alone group. Furthermore, the combination therapy significantly exerted anti-inflammatory and antioxidant effects, as evidenced by reduced serum levels of tumor necrosis factor-alpha (TNF-α), interleukin-1 beta (IL-1β), and malondialdehyde (MDA), alongside increased superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) activities ( p  < 0.05). 16 S rRNA sequencing demonstrated that the combination treatment significantly modulated the gut microbiota composition, decreasing the relative abundance of Proteobacteria from 44% in the infection group to 1.2% ( p  < 0.01), while increasing Firmicutes and Bacteroidetes to levels comparable to healthy controls. Conclusions The research results provides evidence for the therapeutic potential of NG as an effective antibiotic synergist when combined with AMK against avian colibacillosis.

Natural deep eutectic solvent-based simultaneous extraction of aflatoxins and their precursors from edible lotus seeds

Luo Chenghua,Fan Xiaohong,Zhou Yakui,Wu Haifeng,Zhang Lei,Zhao Xiangsheng

The early detection of aflatoxins (AFs) and their biosynthetic precursors is vital for preventing mycotoxin contamination, yet conventional extraction methods generate substantial ecological burdens due to excessive organic solvent consumption. Herein, a tailored menthol-propionic acid Natural Deep Eutectic Solvent (NADES) was developed for the simultaneous extraction of 11 AFs and their precursors from lotus seeds. This refined solvent system demonstrated satisfactory extraction efficiency when coupled with UPLC-MS/MS, and a 50% contamination incidence was observed in real samples. Notably, the NADES-based protocol showed practical advantages under the compared conditions, including over 90% organic solvent reduction and 10~20 times faster extraction than conventional methods. The extraction mechanism was further elucidated at the molecular level through integrated density functional theory (DFT) calculations and molecular dynamics simulations (MDs). Quantum chemical analysis revealed energetically favorable hydrogen bonding and hydrophobic interaction between the NADES system and aromatic rings of AFB 1 , while MDs further demonstrated the formation of persistent hydrogen-bond networks. This work provides a viable, eco-friendly strategy for extracting aflatoxins and their precursors, demonstrating a high potential for applications in the extraction of mycotoxins from food, feed, and herbal products.

High-entropy alloy Janus artificial enzymes for pH-gated sequential redox therapy of drug-resistant bacterial infection

Han Cong,Wang Yongqi,Gao Shihuan,Wang Ting,Du Huili,Long Jie,Tian Weidong,Adeli Mohsen,Cheng Liang,Liu Zhi,Chen Tian,Cheng Chong

Drug-resistant bacterial infections in chronic wounds remain a critical challenge, particularly under persistent inflammation. Here, we report the de novo design of high-entropy alloy (HEA, PtFeCuCoNi)-based Janus artificial enzymes with pH-gated redox biocatalysis for sequential antibacterial and repair functions. The multi-metal synergy stabilizes the d -band center, allowing acidic oxidase/peroxidase-like activity and neutral antioxidase-like activity. In infection, the enzymes generate bactericidal reactive oxygen species (ROS) to eliminate methicillin-resistant Staphylococcus aureus ( MRSA ) and biofilms at ultralow concentrations (8 μg/mL). During healing, they scavenge ROS, alleviate oxidative injury and support cellular proliferation. In MRSA -infected wounds, this dual-action system clears bacteria and then accelerates regeneration through enhanced neovascularization and matrix remodeling. Mechanistic analyses reveal PFKFB3 -mediated metabolic reprogramming, suppression of pro-inflammatory cytokines, and macrophage polarization toward the M2 phenotype. Integrating pH-gated antimicrobial and immunomodulatory repair within one nanoplatform, this strategy addresses the conflicting demands of infection control and tissue healing.

A novel two-channel ratiometric fluorescent probe for monitoring hypochlorous acid fluctuations in inflammation

Chen Yuping,Ma Yan,Wu Lei,Wang Peng,Si Jingxing

Hypochlorous acid (HClO), as an important member of reactive oxygen species (ROS), is associated with various diseases, including cancer progression and inflammation. Monitoring the dynamic changes of HClO in physiological and pathological processes is significant for the diagnosis of these diseases. Herein, we designed and developed a ratiometric fluorescent probe TCF-MB for the specific detection of HClO. The probe features tricyanofuran (TCF) as a strong electron-withdrawing group, a benzene ring, and a carbon-carbon double bond as the π-conjugation bridge, and n -butyl-substituted phenothiazine as the electron donor and reactive site. TCF-MB exhibits a pronounced intramolecular charge transfer (ICT) effect. After the addition of HClO, the sulfur atom in TCF-MB is oxidized to sulfoxide, resulting in the blue shift of absorption and emission spectra due to the interruption of the ICT effect. TCF-MB displayed excellent sensitivity and selectivity toward HClO with the LOD of 0.23 μM. Furthermore, TCF-MB can effectively monitor both endogenous and exogenous HClO in living cells and inflammatory zebrafish with low cytotoxicity. These results demonstrated that probe TCF-MB could be a robust tool for tracking HClO dynamics in physiological and pathological environments. Graphical

Enhancing the electrochemical performance of VPO5 anode through direct incorporation of CNTs via ball-milling

Zhao Mingjie,Wen Weidong,Lu Bingxu,Zhang Junpu,Wang Huanwen,Gong Yansheng,Jin Jun,Liu Yanqi,Wang Rui

VPO 5 is another anode material with a relatively high lithium intercalation/deintercalation potential plateau, which is less prone to lithium dendrite formation during cycling and boasts superior safety performance. However, its low Coulombic efficiency and poor electrical conductivity restrict the enhancement of its overall electrochemical performance. In this research, carbon nanotubes are introduced to modify the VPO 5 anode via a simple and scalable ball-milling method. Results show that the composite material presents an enhanced conductivity and specific surface area, thus improves the electrochemical performance. The optimized material retains a capacity retention rate of 93.3% after 350 cycles and delivers a specific capacity of 241.2 mAh g − 1 at a rate of 10 C, which represents a significant improvement compared with the unmodified pristine VPO 5 . This study proposes a feasible strategy for modifying metal phosphate anode materials, and further provides guidance for the development of next-generation batteries with high specific energy and safety.

Preparation and performance of platinum catalysts for hydrosilylation supported by polyphenols modified SBA-15

Sun Zepan,Hu Tao,Pan Yajing,Liu Zhiqing,Wu Chonggang

Surface modification of porous particles with functional groups is a common way to anchor noble metal catalysts. Herein, surface modification of mesoporous silica (SBA-15) was performed using polyphenols (PNs) through oxidative copolymerization of tea polyphenols and theophylline. The PNs modification ratios on subsequent both Pt loading and chemical states of supported Pt are thoroughly investigated. Catalytic activity of the resulting catalysts was evaluated through various hydrosilylation reactions, while its recyclability was also examined. Results show that modification with PNs can not only facilitates the loading of Pt, but also favors the formation of more active Pt 0 species, thereby increase the catalytic activity of the catalyst. Among them, the SBA-15-PNs-Pt-2 catalyst with the highest PNs ratios exhibits both high activity, good selectivity of anti-Markovnikov addition products and excellent recycling performance. Meanwhile, whether it is under low catalyst loadings or toward the hydrosilylation reactions between a series of substrates, the SBA-15-PNs-Pt-2 catalyst all exhibit undeclined activity, which again signifies the substantial role of PNs in both loading and anchoring Pt species.

Washable, low-cytotoxicity, and antibacterial CuO nanoparticle-coated cotton fabrics prepared via in situ thermal oxidation of a copper(II)-hyperbranched poly(amide–amine) complex

Cai Shenao,Yang Yurui,Yao Lirong,Xu Sijun,Zhang Chengjiao

Traditional copper oxide nanoparticles (CuONPs) for antimicrobial textiles face significant challenges. First, the synthesis is reagent-intensive, requiring hazardous substances such as sodium hydroxide. Second, the resulting CuONPs usually exhibit poor aqueous dispersibility and low affinity for fabrics. Herein, an amino-terminal hyperbranched poly(amide–amine) and Cu 2+ complex (HBPAA-Cu 2+ ) was developed. This complex can be coated to cotton fabric via a conventional dipping-rolling-drying process and converted in situ into HBPAA-capped CuONPs (HBPAA-CuO) through thermal oxidation. This approach avoided the need for additional alkaline precipitants and surfactants during the formation of CuO and was expected to reduce the amount of CuONP-containing wastewater generated compared to conventional wet-chemical synthesis routes. Owing to the combined effect between the antimicrobial cationic HBPAA and the CuONPs, no colonies of E. coli and S. aureus were detected for the treated fabric even after 50 laundering cycles (Cu content > 100 mg/kg). Furthermore, cell viability remained above 80% with Cu content below 300 mg/kg, indicating low cytotoxicity. The treatment also preserved the fabric’s original wearability, including tensile strength, air permeability, and flexural stiffness. This strategy offers a facile and sustainable pathway for the large-scale production of washable, low-cytotoxicity, and antimicrobial textiles.

Vertically oriented 1D/3D heterojunction for efficient and stable inverted perovskite solar cells

Hongyu Chen,Xueliang Zhu,Kangwei Mo,Ning Ma,Shuting Pang,Bianyuan Ren,Xinyu Jiang,Man Yang,Yibo Tu,Guodong Li,Mingzhou Yu,Wensheng Yan,Gongxun Bai

1D/3D heterostructure perovskite solar cells have attracted considerable attention owing to their exceptional stability. However, conventional 1D perovskites typically align parallel to the substrate or in disordered orientations, hindering longitudinal carrier transport and limiting device efficiency. Here, we introduce a bilayer interface engineering strategy to induce a vertically oriented 1D capping layer on the surface of the 3D perovskite surface. This vertically ordered 1D/3D heterojunction enables extended carrier lifetime with rapid interlayer charge transport. More importantly, the vertically oriented 1D capping layer not only tunes energy level alignment but also significantly reduces interface trap density, thereby minimizing photovoltage deficit to 350 mV. With these merits, the champion device achieves a maximum power conversion efficiency of 25.9% (26.0% certified). Under high-temperature 85 °C and continuous illumination conditions, the 1D/3D devices maintain 83% and 86% of their initial efficiency after 500 and 1200 hours, respectively, demonstrating their excellent practical durability. Disordered orientations of the 1D component limit carrier transport in 1D/3D heterostructures. Chen et al. report bilayer interface engineering to induce vertically ordered 1D capping layers, enhancing charge transport, and achieving a certified efficiency of 26.0% with strong thermal and light stability.

Single-particle atomic-scale strain-gradient engineering for high-performance fuel cells

Zhao Dafu,Tang Zisheng,Liu Jinfeng,Hu Zhiyi,Yin Zhiwen,Lv Jieheng,Liao Xiaobin,Wang Xiaoqian,Liu Yingfei,Liu Damin,Chen Lihua,Su Bao-Lian,Zhao Dongyuan,Liu Yong

Practical electrochemical energy conversion requires electrocatalysts that coordinate multiple elementary steps at spatially distinct active sites, yet atomic-level control of such site-specific reactivity within a single heterogeneous particle remains challenging. Here we propose and realize atomic-scale gradient strain as a design concept for heterogeneous electrocatalysis. Using Pd@Pt core-shell tetrahedra as a model system, we construct a continuous lattice-strain gradient across individual 3–4 atomic-layer Pt {111} epitaxial shells, where lattice-mismatch-driven compression relaxes from edges to center regions. This single-particle strain gradient, spanning approximately −8% to −2%, spatially links *O 2 activation at highly compressed sites with *OH weakening at moderately compressed sites through kinetically accessible intermediate redistribution. The catalysts exhibit competitive oxygen reduction reaction performance, with mass and specific activities of 2.19 A mg Pt ⁻ 1 and 3.01 mA cm⁻ 2 at 0.9 V vs reversible hydrogen electrode, while retaining 91% activity after 20 k cycles. In membrane electrode assemblies, they achieve 0.57 A mg Pt ⁻ 1 , and peak power densities of 2.10 W cm⁻ 2 in H 2  − O 2 and 1.16 W cm⁻ 2 in H 2 −air, with over 90% performance retention after 20 k cycles.

Stretchable capacitor with patterned structure by microfluidic molding technology

Feng Pengdong,Li Yang,Liu Yanran,Shi Yonghong

Stretchable capacitors are vital for flexible wearables due to outstanding mechanical adaptability and electrical stability. Herein, a microfluidic route is proposed to fabricate patterned stretchable capacitive sensors with silicone rubber–based nanocomposites. A detachable microfluidic mold efficiently patterns conductive and dielectric layers. The regulation effect of (Koch curve and rhombus grid curve) structures on stretchability is analyzed. Patterns limit conductive composite strain below 32% for stable conductance. Dielectrics stretch evenly above 100% without fracture. These features jointly deliver excellent stretchability and electrical performance. Tests show the patterned devices possess great capacitance-strain linearity. The composite dielectric material of (PDMS + AgNPs + Ni) achieves linearity of 0.968 within 0–50% strain. Sensors with (Ecoflex + MWCNT) dielectrics display reverse sensing behavior unlike prior studies. The devices gain R 2  = 0.995 at 0–100% strain to show excellent linearity and reliability. The composite dielectrics and microfluidic patterning strategy have bright prospects in flexible electronics and wearable sensing.
