Polydopamine-Coated Magnetic Composite Particles with an Enhanced Photothermal Effect

Rui Zheng,Sheng Wang,Ye Tian,Xinguo Jiang,Deliang Fu,Shun Shen,Wuli Yang

Recently, photothermal therapy (PTT) that utilizes photothermal conversion (PTC) agents to ablate cancer under near-infrared (NIR) irradiation has attracted a growing amount of attention because of its excellent therapeutic efficacy and improved target selectivity. Therefore, exploring novel PTC agents with an outstanding photothermal effect is a current research focus. Herein, we reported a polydopamine-coated magnetic composite particle with an enhanced PTC effect, which was synthesized simply through coating polydopamine (PDA) on the surface of magnetic Fe3O4 particles. Compared with magnetic Fe3O4 particles and PDA nanospheres, the core–shell nanomaterials exhibited an increased NIR absorption, and thus, an enhanced photothermal effect was obtained. We demonstrated the in vitro and in vivo effects of the photothermal therapy using our composite particles and their ability as a contrast agent in the T2-weighted magnetic resonance imaging. These results indicated that the multifunctional composite particles with enhanced photothermal effect are superior to magnetic Fe3O4 particles and PDA nanospheres alone.

Synthesis of Au@MOF core–shell hybrids for enhanced photodynamic/photothermal therapy

Xue Cai,Yuewu Zhao,Li Wang,Mingchao Hu,Zeen Wu,Lifen Liu,Weipei Zhu,Renjun Pei

Photodynamic/photothermal therapy (PDT/PTT) has become a research focus of cancer treatment due to the non-invasiveness, spatio-temporal controllability, and effectiveness of repeated treatment. Here, Au@MOF core–shell hybrids were designed and constructed by the layer-by-layer method, and the thickness of the MOF shell can be adjusted by controlling the coordination reaction between the layers. Au nanorod cores mainly produce the PTT effect due to their strong absorbance at 650 nm. The porphyrin ligand in the MOF shell can convert O2 into 1O2 under light conditions, resulting in a high PDT effect. Moreover, the metal node Fe3O(OAc)6(H2O)3+ cluster of the MOF can catalyze the decomposition of H2O2 into O2 to overcome the hypoxic environment of tumors, which further improves the effect of PDT. The combination of the porphyrin ligand in the MOF structure and Au nanorods has promoted the synergistic effects of PDT/PTT. As expected, the results confirmed that Au@MOF hybrids showed no obvious biotoxicity in both cells and animal experiments, and exhibited good biocompatibility. With the synergistic effects of PDT/PTT, cancer cells could be effectively killed and tumor growth could be inhibited. In addition, the modification of folic acid on the surface of Au@MOF can further enrich the hybrids at the tumor site and enhance the inhibitory effect on tumors. These studies have proved that PDT and PTT can be effectively combined and have greater advantages in enhancing the treatment of tumors.

Photothermal-augmented reactive oxygen species oxidative synergistic therapy based on nanoceria doped mesoporous polydopamine nanoplatform

Suhua Jiang,Fukai Zhu,Yang Lin,Liu Ran,Fulin Lin,Haina Tian,Ting Hu,Peiyuan Wang,Canzhong Lu

Owing to residual tumor tissues around the treatment margins, it is difficult to totally ablate large tumors, eliminate disseminated and metastatic nodules as well as prevent the tumor recurrence simultaneously by conventional photothermal therapy (PTT). In this work, PTT combined reactive oxygen species oxidative therapy (ROT) was conducted via mesoporous polydopamine nanosphere with nanoceria doping (MPDA-Ce). After surface anchoring with Arg-Gly-Asp (RGD) molecules, the dispersive stability and tumor targeting of this innovative nanoplatform were substantially boosted. In vitro studies revealed that the unique nanotherapeutics had satisfactory photothermal conversion efficiency (η = 51.41%), continuous nanoceria release in cytoplasm of tumor cells after acid microenvironment (pH = 5.5) induced biodegradation. Significantly, hyperthermia amplified the abundant reactive oxygen species generation under 808 nm laser irradiation that potent ROT was realized with only 27.1% cells alive. Strikingly, Indocyanine green (ICG) loaded MPDA-Ce with RGD modification shows robust tumor targeting capability in breast carcinoma bearing Balb/C mice under fluorescent imaging in the second near-infrared window. The combined treatments resulted in complete tumor cells eradication under 808 nm laser illumination both in vitro and in vivo . This work highlights the promising application of MPDA-Ce as a powerful nanoplatform for ROT and PTT combined treatment in clinical.

Coordination between anti-inflammation and antitumor actions for systematic tumor treatments with improved prognosis

Wen-Long Liu,Tao Liu,A-Min Cao,Si-Yong Qin,Jun Feng,Xian-Zheng Zhang

Tumor relapse and cancerometastasis frequently occur post tumor treatments, leading to poor prognosis with high mortality. Unfortunately, therapy-exacerbated inflammation is among the chief causes of poor prognosis, especially for photothermal therapy (PTT). To overcome this mismatch between therapy and prognosis, we design a built-in Pt/polydopamine(PDA) nanoconstruction (PNP) that makes the anti-inflammation potency and therapy outcome positively inter-linked by virtue of their temperature dependency. PDA not only works as the PTT agent but also provides a self-reductive nano-substrate (∼150 nm) for the in-situ generation of ultrasmall Pt nanodots (∼4 nm). The embedding of Pt nanodots affects insignificantly the PTT potency of PDA while importing the enzyme-like ROS-scavenging function to defend inflammation in a therapy-enhanced manner. Specifically, PNP exhibits the increasing anti-inflammation effect with the elevated temperature caused by PTT, which enables the anti-inflammation pathway to match perfectly PTT modality. In vivo results indicate that PNP leads to better antitumor performance and effective inhibition of tumor relapse and cancerometastasis. This study provides new ideas for systematical tumor treatments with improved prognosis by coordinating anti-inflammation and antitumor actions harmoniously.

Ultrathin Transition Metal Chalcogenide Nanosheets Synthesized via Topotactic Transformation for Effective Cancer Theranostics

Jingjing Wu,Shaomin Zhang,Xuan Mei,Ning Liu,Tingting Hu,Ruizheng Liang,Dan Yan,Min Wei

Ultrathin transition metal chalcogenide (TMC) nanosheets with ultrahigh photothermal conversion efficiency (η) and excellent stability are strongly desired in the application of photothermal therapy (PTT). However, the current synthetic methods of ultrathin TMC nanosheets have issues in obtaining uniform morphology, good dispersion, and satisfactory PTT behavior. Herein, ultrathin nanosheets of CoFe-selenide (CFS) with a finely controlled structure were prepared via a topological structural transformation process from an ultrathin CoFe-layered double hydroxide (LDH) precursor, followed by surface modification with poly(ethylene glycol) (PEG). The as-prepared CFS-PEG nanosheets inherit the ultrathin morphology of CoFe-LDH and exhibit an outstanding photothermal performance with a η of 74.5%, which is the first rank level of reported two-dimensional (2D) TMC nanosheet materials. The CFS-PEG nanosheets possess a satisfactory photoacoustic (PA) imaging capability with an ultralow detection limit (5 ppm) and simultaneously superior T2 magnetic resonance imaging (MRI) performance with a large transverse MR relaxivity value (r2) of 347.7 mM–1 s–1. Moreover, in vitro and in vivo assays verify superior anticancer activity with a dramatic photoinduced cancer cell apoptosis and tumor ablation. Therefore, a successful paradigm is provided for rational design and preparation of ultrathin TMC nanosheets in this work, holding enormous potential in cancer theranostics.

Mild thermotherapy and hyperbaric oxygen enhance sensitivity of TMZ/PSi nanoparticles via decreasing the stemness in glioma

Zeng Xiaofan,Wang Qi,Tan Xuan,Jia Le,Li Yuwei,Hu Mingdi,Zhang Zhijie,Bai Xicheng,Zhu Yanhong,Yang Xiangliang

Background Glioma is a common brain tumor with a high mortality rate. A small population of cells expressing stem-like cell markers in glioma contributes to drug resistance and tumor recurrence. Methods Porous silicon nanoparticles (PSi NPs) as photothermal therapy (PTT) agents loaded with TMZ (TMZ/PSi NPs), was combined with hyperbaric oxygen (HBO) therapy in vitro and in vivo. To further investigate underlying mechanism, we detected the expression of stem-like cell markers and hypoxia related molecules in vitro and in vivo after treatment of TMZ/PSi NPs in combination with PTT and HBO.Results NCH-421K and C6 cells were more sensitive to the combination treatment. Moreover, the expression of stem-like cell markers and hypoxia related molecules were decreased after combination treatment. The in vivo results were in line with in vitro. The combination treatment presents significant antitumor effects in mice bearing C6 tumor compared with the treatment of TMZ, PTT or TMZ/PSi NPs only. Conclusion These results suggested the TMZ/PSi NPs combined with HBO and PTT could be a potential therapeutic strategy for glioma.

Photothermal-driven disassembly of naphthalocyanine nano-photosensitizers for photothermal and photodynamic therapy

Wei Su,Xiantao Jiang,Ying Zhang,Chenxiang Lin,Jun'an Xiao,Peiyuan Li

The disassembly of nanomaterials is of particular interest for high-quality imaging and targeted therapies in the field of nanomedicine. In this study, we developed a novel strategy for fabricating self-assembled naphthalocyanine photosensitizers ( [email protected] ) with intrinsically unique photochemical and photophysical properties. [email protected] could be disassembled under the photothermal effect, and its photoactivity could be enhanced by 780 nm laser irradiation. Moreover, [email protected] generates reactive oxygen species, including superoxide radicals (O 2 • – ) and singlet oxygen ( 1 O 2 ), as well as good photothermal properties, facilitating the application of multifunctional phototherapy. In vitro evaluation indicated that [email protected] possesses an excellent bactericidal effect under a combination of photodynamic (PDT) and photothermal therapy (PTT). The in vivo treatment of a full-layer skin defect model of Escherichia coli ( E. coli ) infection showed that [email protected] had superior antibacterial and wound-healing abilities. These results provide the basis for a feasible strategy to enhance the phototherapeutic effect of photosensitizer (PS) systems.

Black phosphorus quantum dots camouflaged with platelet-osteosarcoma hybrid membrane and doxorubicin for combined therapy of osteosarcoma

Xu Yanlong,Du Longhai,Han Binxu,Wang Yu,Fei Jichang,Xia Kuo,Zhai Yonghua,Yu Zuochong

Background Osteosarcoma (OS) is the most prevalent primary malignant bone tumor. However, single-agent chemotherapy exhibits limited efficacy against OS and often encounters tumor resistance. Therefore, we designed and constructed an integrated treatment strategy of photothermal therapy (PTT) combined with chemotherapy and used a surface-encapsulated platelet-osteosarcoma hybrid membrane (OPM) that enhances circulation time and enables OS-specific targeting. Results The OPM functions as a shell structure, encapsulating multiple drug-loaded nanocores (BPQDs-DOX) and controlling the release rate of doxorubicin (DOX). Moreover, near-infrared light irradiation accelerates the release of DOX, thereby extending circulation time and enabling photostimulation-responsive release. The OPM encapsulation system improves the stability of BPQDs, enhances their photothermal conversion efficiency, and augments PTT efficacy. In vitro and ex vivo experiments demonstrate that BPQDs-DOX@OPM effectively delivers drugs to tumor sites with prolonged circulation time and specific targeting, resulting in superior anti-tumor activity compared to single-agent chemotherapy. Furthermore, these experiments confirm the favorable biosafety profile of BPQDs-DOX@OPM. Conclusions Compared to single-agent chemotherapy, the combined therapy using BPQDs-DOX@OPM offers prolonged circulation time, targeted drug delivery, enhanced anti-tumor activity, and high biosafety, thereby introducing a novel approach for the clinical treatment of OS.

Cu2O-SnO2-PDA heterozygous nanozyme doped hydrogel mediated conglutinant microenvironment regulation for wound healing therapy

Jinghua Li,Pengshan Guo,Shegan Gao,Jianping Wang,Ji Cheng,Wenxuan Fan,Xiaoran Liu,Xiaozhi Zhang,Kun Lei

Bacterial infection significantly hinders the wound healing process. Overuse of antibiotics has led to the rise of drug resistance in bacteria, making the development of smart medical dressings that promote wound healing without antibiotics, a critical need. In this study, Cu₂O-SnO₂-PDA (PCS) nanoenzymes with Fenton-like activity and high photothermal conversion efficiency were developed. These nanoenzymes were then incorporated into a hydrogel through cross-linking of acrylamide (AM) and N-[Tris-(hydroxymethyl)methyl] acrylamide (THMA), forming a tough, highly-adhesive, and self-healing composite hydrogel (AT/PCS) with antimicrobial properties. The AT/PCS hydrogel exhibits excellent mechanical strength and adhesion, facilitating increased oxygen levels and strong adherence to the wound site. Moreover, it effectively regulates the wound microenvironment by combining synergistic chemodynamic therapy (CDT) and photothermal therapy (PTT) for antibacterial treatment. The AT/PCS hydrogel enhances collagen deposition and expedites wound healing in a rat model, largely due to its potent antibacterial properties.

Prodrug-Loaded Zirconium Carbide Nanosheets as a Novel Biophotonic Nanoplatform for Effective Treatment of Cancer

Quan Liu,Zhongjian Xie,Meng Qiu,Inseob Shim,Yunlong Yang,Sisi Xie,Qinhe Yang,Dou Wang,Shiyou Chen,Taojian Fan,Bo Ding,Ziheng Guo,Dickson Adah,Xinhuang Yao,Yuhua Zhang,Hong Wu,Zongze Wu,Chaoying Wei,Hongzhong Wang,Hyeong Seok Kim,Qingshuang Zou,Qiaoting Yan,Zhen Cai,Jong Seung Kim,Li-Ping Liu,Han Zhang,Yihai Cao

Conventional chemotherapy and photothermal therapy (PTT) face many major challenges, including systemic toxicity, low bioavailability, ineffective tissue penetration, chemotherapy/hyperthermia-induced inflammation, and tumor angiogenesis. A versatile nanomedicine offers an exciting opportunity to circumvent the abovementioned limitations for their successful translation into clinical practice. Here, a promising biophotonic nanoplatform is developed based on the zirconium carbide (ZrC) nanosheet as a deep PTT-photosensitizer and on-demand designed anticancer prodrug SN38-Nif, which is released and activated by photothermia and tumor-overexpressed esterase. In vitro and in vivo experimental evidence shows the potent anticancer effects of the integrated ZrC@prodrug biophotonic nanoplatform by specifically targeting malignant cells, chemotherapy/hyperthermia-induced tumor inflammation, and angiogenesis. In mouse models, the ZrC@prodrug system markedly inhibits tumor recurrence, metastasis, inflammation and angiogenesis. The findings unravel a promising biophotonic strategy for precision treatment of cancer.

Combination of PEG-decorated black phosphorus nanosheets and immunoadjuvant for photoimmunotherapy of melanoma

Shiyu Wan,Boya Zhang,Sai Li,Bin He,Yuji Pu

Photoimmunotherapy, which combines local photothermal therapy (PTT) with immunological stimulation, is a promising modality for cancer treatment. Herein, we have reported a photothermal-immunotherapy of melanoma using pegylated black phosphorus nanosheets (BP-PEG NSs) and imiquimod (R837) as the photothermal conversion agent and the immunoadjuvant, respectively. The photothermal stability of BP NSs was remarkably enhanced after the modification of poly(ethylene glycol) (PEG) by electrostatic interactions. The in situ generation of tumor-associated antigens by PTT elicited a strong immune response in the presence of R837, achieving a photoimmunotherapy of B16 melanoma. This photoimmunotherapy stimulated a stronger immune response both in vitro and in vivo than monotherapy, inducing a much greater release of cytokines such as IL-6, IL-12, and TNF-α. In vivo antitumor studies in B16 tumor-bearing mice demonstrated that photoimmunotherapy showed the best tumor inhibition effects. Our study suggested that BP-PEG NS-based PTT primed with an immunoadjuvant can be used for synergistic photoimmunotherapy of melanomas.

Synergistic combination: Promising nanoplatform W-POM NCs@ HKUST-1 for photothermal and chemodynamic reinforced anti-tumor therapy

Jiale Liu,Mengyao Huang,Zhongyu Hua,Jiatong Ni,Yi Dong,Zeran Feng,Tiedong Sun,Chunxia Chen

Chemodynamic therapy (CDT) uses the conversion of H 2 O 2 to ·OH triggered by Fenton-like reactions to treat tumors, but the treatment is not complete due to insufficient intracellular H 2 O 2 content. Photothermal therapy (PTT) uses photothermal materials for photothermal conversion to kill tumors, but it is limited by the extent of exogenous light transmission and cannot achieve satisfactory therapeutic effects. Therefore, designing a composite nanomaterial that amplifies CDT and couples it with PTT for tumor treatment is a major challenge. Herein, we designed a W-POM NCs@HKUST-1 nanoplatform that combines PTT and CDT and has better targeting properties. It shows high stability and photothermal conversion efficiency under the irradiation of near-infrared light and efficiently catalyzes the generation of intracellular hydroxyl radicals, this composite material exhibits higher lethality than single therapeutic strategies. This study expands the application area of POM@MOF composite nanomaterials and provides a promising means of enhanced tumor therapy.
