概述

沙黄也叫藏红T，分子式为C₂₀H₁₉ClN₄，分子量为350.85，CAS号为477-73-6。为红棕色粉末，易溶于水成红色溶液，溶于乙醇红色带黄色荧光。对盐酸为蓝红色溶液，过多则呈紫色，大量过多则转为蓝色；对氢氧化钠则生成棕红色沉淀；对硫酸则为绿色溶液，稀释时先变成蓝色，渐变成紫色，最后变成红色。最大吸收波长530nm，有刺激性。

沙黄/藏红T水溶液 (2%) 主要由沙黄和去离子水组成，可用于布氏杆菌染色或其他用途。该产品仅适用于科研实验，不可做他用。

自备材料：

1、载玻片、盖玻片

2、酒精灯

3、光学显微镜

操作步骤（仅供参考）：

1、涂片：载玻片编号，于载玻片中央滴加沙黄/藏红T水溶液，加热至出现蒸汽，约0.5～1min。

2、加盖洁净的盖玻片，轻轻按压制成压片。

3、光学显微镜在低倍、高倍或油镜下观察细菌形态。

注意事项：

1、涂片之前应事先在背面做好圆圈标记，以便判断后续试验的位置。

2、待检细菌培养时间会影响染色，阳性菌培养过长或已死亡或细菌溶解，都常呈阴性反应。

3、为了您的安全和健康，请穿实验服并戴一次性手套操作。

Safranin, also known as Safranin T, has a molecular formula of C₂₀H₁₉ClN₄, a molecular weight of 350.85, and a CAS number of 477-73-6. It is a reddish-brown powder that is soluble in water to form a red solution and soluble in ethanol with a red color and yellow fluorescence. With hydrochloric acid, it forms a blue-red solution, turning purple with excess acid and blue with a large excess. With sodium hydroxide, it produces a reddish-brown precipitate. With sulfuric acid, it forms a green solution that first turns blue, then purple, and finally red upon dilution. Its maximum absorption wavelength is 530nm, and it is irritating.

2% aqueous solution of Safranin/Safranin T consists mainly of safranin and deionized water. It can be used for Brucella staining or other purposes. This product is intended for research experiments only and should not be used for other purposes.

Operating Steps (For Reference Only):

1. Smear Preparation: Label the slide, place a drop of the safranin/safranin T aqueous solution in the center, and heat until steam is observed, approximately 0.5 to 1 minute.

2. Mounting: Place a clean cover slip over the preparation and gently press to make a squash mount.

3. Observation: Examine the bacterial morphology using the optical microscope under low, high, or oil immersion objective.

Precautions:

1. Marking: Prior to smearing, make a circular mark on the back of the slide to identify the area for subsequent examination.

2. Incubation Time: The incubation time of the test bacteria can affect staining results. Over-incubation, death, or lysis of Gram-positive bacteria often result in false-negative reactions.

3. Safety: For your safety and health, wear a lab coat and disposable gloves during the operation.
