固定的目的在于保存细胞和组织的原有形态结构，固定剂能阻止内源性溶酶体酶对自身组织和细胞的自溶、抑制细菌和霉菌的生长，固定剂通过凝固、生成添加化合物等使蛋白质内部结构发生改变，从而使酶失活。固定液分为醛类固定液、汞类固定液、醇类固定液、氧化剂类固定液、苦味酸盐类固定液等，较为常用的是醛类中的福尔马林、醇类中的乙醇。
福尔马林醇固定液主要由乙醇、福尔马林组成，该固定液属于脱水-交联固定剂，在辨认特定抗体的免疫组化反应中能产生极好的结果，也有利于含有多量脂肪的大样本固定。
该试剂仅用于科研领域，不适用于临床诊断或其他用途。
操作步骤（仅供参考）：
1、按实验具体要求操作。
2、一般标本固定时间控制 1-4 h/mm，大多数控制在 4-24 h，大标本应适当延长固定时间。
注意事项：
1、乙醇-福尔马林-醋酸固定液有一定刺激性和腐蚀性，请在通风环境下小心操作。
2、组织取材的厚度不同，固定时间也不同；常规活检组织比较适合的厚度为 2-4 mm，一般不超过 6 mm，对组织恰当的选材有利于固定液的渗透。
3、固定液的容量应足够，一般固定液与组织块的体积比率应大于10：1；如果容积不够大，可以在固定期间更换 1-3 次固定液。
4、温度对固定的影响很明显，提高温度可以加速固定作用，但温度不宜过高。
5、取出新鲜组织后应及时固定，无法及时固定时应保存于生理盐水中及时送检。
6、试剂开封后请尽快使用，以防影响后续实验效果。
The purpose of fixation is to preserve the original morphological structure of cells and tissues. Fixatives can prevent autolysis of tissues and cells by endogenous lysosomal enzymes and inhibit the growth of bacteria and fungi. Fixatives inactivate enzymes by altering the internal structure of proteins through coagulation, formation of addition compounds, etc. Fixatives are divided into aldehyde fixatives, mercuric fixatives, alcohol fixatives, oxidizing agent fixatives, picrate fixatives, etc. The more commonly used ones are formalin among aldehydes and ethanol among alcohols.
Formalin-Alcohol Fixative is mainly composed of ethanol and formalin. This fixative is a dehydration-crosslinking fixative and can produce excellent results in immunohistochemical reactions for identifying specific antibodies. It is also beneficial for the fixation of large specimens containing a large amount of fat.
This reagent is for research use only and is not intended for clinical diagnosis or other purposes.
Procedure (for reference only):
1.Operate according to the specific requirements of the experiment.
2.Generally, the fixation time for specimens should be controlled at 1–4 h/mm, with most controlled at 4–24 h; for large specimens, the fixation time should be appropriately extended.
Precautions:
1.Ethanol-formalin-acetic acid fixative is somewhat irritating and corrosive. Please operate carefully in a ventilated environment.
2.The fixation time varies with the thickness of the tissue sample. The appropriate thickness for routine biopsy tissues is 2–4 mm, generally not exceeding 6 mm. Proper selection of tissue is conducive to the penetration of the fixative.
3.The volume of fixative should be sufficient. Generally, the volume ratio of fixative to tissue block should be greater than 10:1; if the volume is insufficient, the fixative can be replaced 1–3 times during fixation.
4.Temperature has a significant effect on fixation. Increasing the temperature can accelerate fixation, but the temperature should not be too high.
5.After removing fresh tissue, it should be fixed promptly. If timely fixation is not possible, it should be stored in physiological saline and sent for examination promptly.
6.After opening the reagent, please use it as soon as possible to prevent adverse effects on subsequent experimental results.
