We used chondroitinase to selectively degrade various chondroitin sulfate/dermatan sulfate, combined with gel filtration and anion exchange HPLC, to prepare high-purity C4, 5 unsaturated disaccharides standards with various sulfation modes. Including seven common unsaturated disaccharides after chondroitin sulfate/dermatan sulfate degradation by chondroitinase: Δ di-0S (Δ UA - [1 → 3] - GalNAc), Δ di-4S (Δ UA - [1 → 3] - GalNAc-4S), Δ di-6S (Δ UA - [1 → 3] - GalNAc-6S), Δ di-4S6S (Δ UA - [1 → 3] - GalNAc-4S), Δ di-2S4S (Δ UA-2S - [1 → 3] - GalNAc-6S), and Δ di-2S4S6S (Δ UA-2S - [1 → 3] - GalNAc-6S) 3] - GalNAc-4S-6S and its corresponding 2-Aminobenzamide (2AB) fluorescently labeled disaccharide standard
