概述

MCDB 153 培养基是为特定细胞设计的低蛋白和无血清培养的培养基，MCDB 153 是 MCDB 培养基的一种，是 Knedler 和 Ham 设计开发出来的，最初用于培养人微血管内皮细 (HMVEC)。MCDB 153 培养基除用于内皮细胞外，还可用于其它细胞类型，如肝细胞、平滑肌细胞、心肌细胞等。

操作指南：

1、将胰酶（胰蛋白酶 0.25%，EDTA 0.02%) 及完全培养基提前恢复至室温。

2、弃掉细胞培养基，用 DPBS 洗涤细胞单层。

3、加入适当的体积（例如，在 75 cm² 的容器中加入 5 mL）的胰酶，确保胰酶完全覆盖细胞单层，37°C 孵育1-3min，用倒置显微镜观察，确保细胞完全脱离培养瓶表面（轻轻拍打培养瓶有助于细胞脱落）。

4、加入 5-10 mL 完整培养基，冲洗培养瓶壁并混匀，将细胞悬液收集到 15 mL 离心管中。

5、1000 rpm 离心 5-10 min，丢弃上清，用 2-5 mL 完全培养基重悬细胞。

6 、测定活细胞密度和存活率，根据细胞类型及实验需求按常规程序接种，孵育和传代。

注意事项：

1、室温运输，到货后 2-8℃，避光保存，避免反复冻融，有效期1年。

2、本产品已滤菌，使用前轻轻摇匀。

MCDB 153 Medium is a low-protein and serum-free culture medium designed for specific cell types. MCDB 153 is one of the MCDB series media, designed and developed by Knedler and Ham. It was initially applied to the culture of human microvascular endothelial cells (HMVEC). In addition to endothelial cells, MCDB 153 medium is also suitable for other cell types, such as hepatocytes, smooth muscle cells, cardiomyocytes and so on.

Product Features:

MCDB 153 medium is developed for the serum-free culture of human keratinocytes. Before use, additives required for the growth of human keratinocytes need to be supplemented, such as recombinant epidermal growth factor (rEGF), bovine pituitary extract (BPE), hydrocortisone, etc.

Operating Guidelines:

1. Warm trypsin (0.25% trypsin, 0.02% EDTA) and complete medium to room temperature in advance.

2. Discard the spent culture medium and rinse the cell monolayer with DPBS.

3. Add an appropriate volume of trypsin (for example, 5 mL for a 75 cm² culture vessel) to fully cover the cell monolayer. Incubate at 37 °C for 1-3 minutes. Observe under an inverted microscope until cells are completely detached from the surface of the culture flask. Gently tapping the flask can facilitate cell detachment.

4. Add 5-10 mL of complete medium, rinse the inner wall of the culture flask and mix well. Collect the cell suspension into a 15 mL centrifuge tube.5

5. Centrifuge at 1000 rpm for 5-10 minutes, discard the supernatant, and resuspend the cells with 2–5 mL of complete medium

6. Determine viable cell density and cell survival rate. Perform cell seeding, incubation and subculture following standard procedures according to cell type and experimental requirements.

Precautions:

1. Transport at room temperature. Upon receipt, store at 2- 8 °C in the dark and avoid repeated freeze-thaw cycles. The shelf life is 1 year.

2. This product is sterile filtered; gently invert and mix well before use.

3. Our company shall not be liable for any accidents caused by improper use of this product.
