概述

甘油PBS封片剂是一种用于常规封固剂，亦可用于减缓荧光淬灭的封固，可以用于减缓各种常见荧光染料的荧光淬灭，主要由甘油、PBS缓冲液组成；对于减缓荧光淬灭，其效果不如抗荧光淬灭封片剂 (Antifade Mounting Medium)，按每个样本需要50-100μl封片剂计算，5ml可以用于至少50样本的封片。该试剂仅用于科研领域，不适用于临床诊断或其他用途。

自备材料：

1、载玻片

2、盖玻片

3、荧光显微镜

操作步骤(仅供参考)：

1、贴壁细胞样本

①染色完成后，吸去染色液。

②滴1滴甘油PBS封片剂于载玻片上，盖上贴有细胞的盖玻片，让细胞接触封片液，尽量避免气泡。

③在荧光显微镜下观察。

2、组织切片

①染色完成后，吸去染色液。

②滴1滴甘油PBS封片剂于组织切片上，盖上盖玻片，让切片接触封片液，尽量避免气泡。

③在荧光显微镜下观察组织切片。

3、其它样品

其它样品参考细胞样品或组织切片进行操作。

注意事项：

1、荧光物质均易发生淬灭，染色后的样品宜避光保存。

2、在使用甘油PBS封片剂的情况下，虽然可减缓淬灭，仍需尽量避光。

3、为了您的安全和健康，请穿实验服并戴一次性手套操作。

4、试剂开封后请尽快使用，以防影响后续实验效果。

Product Introduction

Glycerol-PBS Mounting Medium is a general-purpose mounting agent that can also be used to mitigate fluorescence quenching of various common fluorescent dyes. It is mainly composed of glycerol and PBS buffer. Its performance in reducing fluorescence quenching is inferior to that of Antifade Mounting Medium. Based on the dosage of 50–100 μl per sample, 5 ml of the medium can be used for mounting at least 50 samples. This reagent is intended for research use only and not for clinical diagnosis or other purposes.

Materials to Be Prepared by Users

1. Glass slides

2. Coverslips

3. Fluorescence microscope

Operating Procedures (For Reference Only)

1. Adherent cell samples

① After staining, aspirate the staining solution.

② Place 1 drop of Glycerol-PBS Mounting Medium onto the glass slide, then cover it with the coverslip with adherent cells attached, ensuring the cells come into contact with the mounting medium and minimizing air bubbles as much as possible.

③ Observe under a fluorescence microscope.

2. Tissue sections

① After staining, aspirate the staining solution.

② Place 1 drop of Glycerol-PBS Mounting Medium onto the tissue section, then cover it with a coverslip, ensuring the section is in contact with the mounting medium and avoiding air bubbles as much as possible.

③ Observe the tissue section under a fluorescence microscope.

3. Other samples

For other samples, follow the procedures for cell samples or tissue sections.

Precautions

1. All fluorescent substances are prone to quenching; stained samples should be stored away from light.

2. Although Glycerol-PBS Mounting Medium can reduce quenching, samples mounted with it should still be protected from light as much as possible.

3. For your safety and health, wear a lab coat and disposable gloves during operation.

4. Use the reagent as soon as possible after opening to avoid compromising subsequent experimental results.
