概述
尿激酶是从健康男性尿中分离获得的一种专一性很强的碱性蛋白酶，分子量主要由33000和54000两种组成，等电点为PH8～9。尿激酶的提取过程由尿液经过硅胶吸附、硫酸铵沉淀、过滤、层析纯化、除菌、冷冻干燥等步骤制成的白色粉末，易溶于水，是一种专一性很强的碱性蛋白水解酶。
尿激酶制剂为酶类溶血栓药，能激活体内纤溶酶原转为纤溶酶，从而水解纤维蛋白使新鲜形成的血栓溶解。临床主要用于急性心肌梗塞、急性脑血栓形成和脑血管栓塞、肢体周围动静脉血栓、中央视网膜动静脉血栓及其他新鲜血栓闭塞性疾病。亦适用于治疗脑血栓形成、周围血管栓塞、中央视网膜血管栓塞、急性心肌梗死等新鲜血栓栓塞性疾病，以及肾移植、整形外科手术等出现的血栓形成。
Urokinase is a highly specific alkaline protease isolated from the urine of healthy men. The molecular weight is mainly composed of 33000 and 54000 whose isoelectric point is PH 8 ~ 9. Urokinase powder made by the steps of extraction process of urine after silica gel adsorption, ammonium sulfate precipitation, filtration, Chromatography purification, sterilization, freeze drying , soluble in water, alkaline protease is a highly specific.
Urokinase is a kind of enzyme thrombolytic agent, which can activate plasminogen into plasmin, which can hydrolyze fibrin to form thrombus. It is mainly used in the treatment of acute myocardial infarction, acute cerebral thrombosis and cerebral embolism, peripheral arterial and venous thrombosis, central retinal artery and vein thrombosis and other fresh thrombotic occlusive diseases. For formation, peripheral vascular embolism, central retinal vein embolism, acute myocardial infarction and other fresh thromboembolic disease in the treatment of cerebral thrombosis, and renal transplantation, plastic surgery and thrombosis.

生化机理
Specifically cleaves the zymogen plasminogen to form the active enzyme plasmin. Catalytic activity: Specific cleavage of Arg-|-Val bond in plasminogen to form plasmin. Activity regulation: Inhibited by SERPINA5. Inhibited by SERPINE1.
Post-translational: Phosphorylation of Ser-158 and Ser-323 abolishes proadhesive ability but does not interfere with receptor binding. Produced as an inactive single-chain protein (pro-uPA or sc-uPA), is processed into the active disulfide-linked two-chain form of PLAU/uPA by a proteolytic event mediated, at least, by TMPRSS4.
