Rabbit anti Human Caspase-10/CASP-10 Antibody, Recombinant, could be used for WB, ICC, IF and so on. Application: WB: 1/500-1/1000 ICC/IF: 1/50-1/200 Protein Function: Involved in the activation cascade of caspases responsible for apoptosis execution. Recruited to both Fas- and TNFR-1 receptors in a FADD dependent manner. May participate in the granzyme B apoptotic pathways. Cleaves and activates caspase-3, -4, -6, -7, -8, and -9. Hydrolyzes the small- molecule substrates, Tyr-Val-Ala-Asp- -AMC and Asp-Glu-Val-Asp- -AMC. Isoform C is proteolytically inactive.
Recombinant Caspase-10/CASP-10 Antibody (Ab092983) - Western Blot All lanes: Recombinant Caspase-10/CASP-10 Antibody (Ab092983) at 1/1000 dilution Samples: Lysates at 20 µg per lane Secondary: Goat Anti-Rabbit IgG H&L (HRP) (Ab170144) at 1/20000 dilution
Predicted band size: 59 kDa Observed band size: 47, 64 kDa The molecular weight of CASP10 observed on Western blots may vary depending on which of its isoform, truncation, or cleavage forms are present. Full-length CASP10 has a propeptide with 219 amino acid and 6 isoforms produced by alternative splicing. During the processing of Caspase-10, the initial cleavage produces two intermediate products, namely p47 and p43. These intermediates are markers of Caspase-10 activation and can typically be observed 15-30 minutes after cells are stimulated by CD95L or TRAIL. Specifically, the cleavage of Caspase-10 occurs at specific amino acid sites, such as D219 and D372, leading to the release of active subunits (p23/p17 and p12) (PMID: 12198154). Exposure time: 17.8 s
