293T (HEK-293T) cells are an immortalized cell line derived from modified human embryonic kidney cells. They are characterized by high transfection efficiency, rapid proliferation, and the ability to be cultured at high density, making them a core tool cell line for lentiviral/adenoviral packaging, recombinant protein expression, plasmid transfection, gene function verification, and cell model construction. These cells are extremely sensitive to serum quality. Common fetal bovine serum often has issues such as interference from miscellaneous proteins, high endotoxin levels, imbalanced growth factor ratios, and trace exogenous contamination, which can easily lead to loose adherence, clumping and detachment, decreased transfection efficiency, low viral titers, and poor batch-to-batch experimental reproducibility.
This product is a premium fetal bovine serum exclusively customized for 293T cells. It is sourced from healthy fetal cattle aged 6–8 months from compliant pastures, and produced using a dedicated, refined manufacturing process that optimizes the ratios of growth factors and adhesion factors, while strictly controlling endotoxins, hemolytic impurities, exogenous contaminants, and protein precipitates. It is maximally adapted to meet the core requirements of 293T cell adherence, high-density expansion, transient transfection, and viral packaging.
Product Advantages
1.Cell-specific adaptability: Optimized pro-adhesion and pro-proliferation active components address issues such as easy detachment, weak growth, and rapid passage-induced senescence in 293T cells, resulting in regular cell morphology and good refractility.
2.High transfection and high titer compatibility: Low endotoxin and low background of miscellaneous proteins significantly reduce cellular stress responses, markedly improve plasmid transfection efficiency and lentiviral/adenoviral packaging titers, and ensure stable protein expression yields.
3.Extremely low impurity interference: Multi-stage hemolysis removal, lipoprotein removal, and microparticle removal processes result in no significant flocculent precipitates after freeze-thaw cycles, avoiding contamination of culture media and interference with cell observation and subsequent experiments.
4.High safety and freedom from exogenous contamination: Rigorous screening for viruses, mycoplasma, bacteria, and fungi, with no bacteriophages or hidden exogenous contaminants, making it suitable for virology research and genetic engineering studies.
5.High batch-to-batch consistency: Each batch is functionally validated with 293T cells, with strict control over proliferation rate, transfection efficiency, and cell viability, completely avoiding experimental failures caused by batch-to-batch variations common in regular sera.
Production Process Overview
1.This product is manufactured using a standardized process that integrates source selection, low-temperature refining, multi-stage impurity removal, nano-sterilization, and cell-specific validation. The entire process is carried out in Grade C clean areas with Grade A aseptic filling. The core steps are as follows:
2.Raw material screening: Aseptic blood collection from healthy cattle in non-epidemic regions, transported via low-temperature closed cold chain with strict control over hemolysis levels. Raw materials with high impurities or high endotoxin are rejected to ensure serum quality from the source.
3.Low-temperature gradient centrifugation refining: Multi-stage low-temperature high-speed centrifugation thoroughly removes blood cell debris, fibrin, lipoprotein precipitates, and lipid microparticles, reducing the background of miscellaneous serum proteins and minimizing impurity interference in 293T cell culture.
4.Active component preservation: The entire process is carried out at low temperature without heat inactivation or chemical modification, fully preserving core active components such as EGF, FGF, adhesion proteins, and growth hormones to support rapid cell adherence and proliferation.
5.Multi-stage nano-sterile filtration: Terminal filtration with 0.1 μm pore size sequentially retains bacteria, fungi, mycoplasma, and microparticles, achieving sterile, high-purity quality.
6.Cell-specific functional validation and release: All finished products must pass functional tests on 293T cells, including serial passage culture, proliferation rate, transfection efficiency, and viral packaging performance, before batch release.
Scope of Application
Specifically designed for HEK-293T cell in vitro adherent/suspension culture, serial passage expansion, transient transfection experiments, lentiviral/adenoviral/AAV packaging, recombinant protein expression, gene silencing/overexpression functional validation, and drug screening cell model construction. It is also compatible with routine engineering cell lines such as 293, CHO, and Vero.
Usage Instructions
1.Thawing method: Serum stored at –20°C should preferably be thawed overnight at 4°C in a refrigerator, with gentle inversion mixing during the process. After complete thawing, warm in a 37°C water bath. Avoid prolonged exposure at room temperature and avoid repeated freeze-thaw cycles.
2.Medium preparation: Recommended formulation: High-glucose DMEM medium + 10% of this serum + 1% antibiotics (optional). Mix thoroughly before use for 293T cell culture.
3.Culture conditions: Routine culture in a 37°C, 5% CO₂, humidified incubator, with medium changes and passaging as needed.
Note: A small amount of flocculent precipitate after thawing is normal protein precipitation and does not affect use. Do not filter the serum yourself, as this may deplete active components.
Precautions
1.This product is for in vitro cell research use only. It is prohibited for human, clinical, or in vivo injection applications.
2.Trace white flocculent precipitate after thawing is natural protein precipitation and is a normal phenomenon that does not affect cell growth or experimental results.
3.Avoid mixing with low-grade conventional sera to prevent the introduction of impurities, endotoxins, and exogenous contaminants that could affect 293T cell transfection and viral experiments.
4.Maintain a sterile environment throughout all operations to prevent microbial contamination. Use promptly after opening.
5.No acclimation is required when switching between batches; this product offers high batch stability and can be directly substituted.
Disclaimer
This product is for research or industrial use only and is not suitable for clinical therapy. Users should validate the suitability of the serum for their own experimental systems. The manufacturer assumes no liability for deviations in experimental results caused by improper handling
